目的探讨逍遥散降低雌性兔膝关节软骨细胞凋亡的可能作用机制。方法 40只新西兰兔通过手术方法进行膝关节失稳型造模,1周后随机分为中药组、西药组、混合组和对照组各10只,驱赶行走4周,分别灌药或灌生理盐水8周。造模后12周末处理动物,采用免疫组化法检测兔膝关节软骨细胞凋亡基因Caspase-3与细胞色素C的表达。结果中药组、西药组和混合组Caspase-3与细胞色素的C表达均低于对照组,比较有统计学意义;中药组、西药组和混合组之间的比较无统计学意义。结论逍遥散可明显下调Caspase-3与细胞色素的C的表达,可能是逍遥散降低兔膝关节软骨细胞凋亡的机制之一。
Objective To investigate the possible mechanism of Xiaoyaosan in reducing the apoptosis of chondrocytes in knee joint of female rabbits. Methods Forty New Zealand rabbits were surgically performed knee instability modeling. One week later, 10 New Zealand rabbits were randomly divided into TCM group, western medicine group, mixed group and control group, each of which was driven for 4 weeks. The rabbits were drenched with saline or saline 8 weeks. At the end of the 12th week after model establishment, the expression of Caspase-3 and Cyt C in rabbits articular cartilage cells were detected by immunohistochemistry. Results The expressions of Caspase-3 and cytochrome C in Chinese medicine group, western medicine group and mixed group were lower than those in control group, which were statistically significant. There was no significant difference between Chinese medicine group, western medicine group and mixed group. Conclusion Xiaoyaosan can obviously down-regulate the expression of Caspase-3 and cytochrome C, which may be one of the mechanisms that Xiaoyaosan can reduce the apoptosis of rabbit articular cartilage.
