目的探讨气虚病理状态对皮肤吸收和氮酮促透功能的影响。方法将正常家兔皮肤分为2组,A组为药物组,B组为药物加1%氮酮组,气虚家兔皮肤分为4组,C组为气虚皮肤药物组,D组为药物加1%氮酮组,E组为药物加2%氮酮,F组为药物加4%氮酮组,采用双室透皮扩散装置进行茶碱体外经皮渗透实验,分别于2 h、4h、8 h、12 h和24 h取样,采用紫光光度计检测茶碱。计算累积渗透量(Q)、渗透系数(Kp)、稳态流量(Jss)、滞后时间(Tlag)和增渗倍数(ER),进行统计分析。结果 B组4 h、8 h、12 h和24 h累积渗透量均高于A组(t=5.65,P<0.01;t=7.21,P<0.01;t=8.66,P<0.01;t=7.23,P<0.01);C组2 h、4 h、8 h、12 h和24 h累积渗透量均高于A组(t=2.63,P<0.01;t=3.19,P<0.01;t=3.34,P<0.01;t=3.46,P<0.01;t=3.07,P<0.01);D组2 h、4h、8 h、12 h和24 h累积渗透量均高于C组(t=2.98,P<0.01;t=5.13,P<0.01;t=9.50,P<0.01;t=12.74,P<0.01;t=20.98,P<0.01),累积渗透量为E组>D组>F组>C组;C组和A组在JSS和Kp、Tlag差异均无统计学意义(P>0.05);D组、E组和F组Jss和Kp均高于C组(P<0.01)。结论气虚大鼠皮肤对药物的通透性增加,2%氮酮在气虚家兔皮肤的促透效果最佳。
Objective To investigate the effect of pathological condition of qi deficiency on skin absorption and penetration of azone. Methods Normal rabbits were divided into 2 groups: A group was drug group, B group was drug plus 1% Azone group, Qi deficiency skin was divided into 4 groups, C group was Qi deficiency skin drug group, D group was drug plus 1% Azone group, Group E as drug plus 2% Azone, Group F as drug plus 4% Azone as group. The percutaneous permeation of theophylline in vitro was performed by two-compartment transdermal diffusion device at 2 h, 4 h, Samples were taken at 8 h, 12 h and 24 h, and theophylline was detected by violet photometer. The cumulative permeation (Q), permeability (Kp), steady flow (Jss), lag time (Tlag) and fold increase (ER) were calculated and analyzed statistically. Results The cumulative amount of permeation at 4 h, 8 h, 12 h and 24 h in group B was significantly higher than that in group A (t = 5.65, P <0.01; t = 7.21, P <0.01; , P <0.01). The cumulative osmolality of group C at 2 h, 4 h, 8 h, 12 h and 24 h was higher than that of group A (t = 2.63, P <0.01; (P <0.01); t = 3.46, P <0.01; t = 3.07, P <0.01). The cumulative permeation volume in group D at 2 h, 4 h, 8 h, 12 h and 24 h was higher than that in group C (t = P <0.01; t = 5.13, P <0.01; t = 9.50, P <0.01; t = 12.74, P <0.01; t = 20.98, There was no significant difference between JS and Kp and Tlag in group C and group A (P> 0.05). Jss and Kp in group D, group E and group F were higher than those in group C (P <0.01). Conclusion The permeability of drug on the skin of Qi deficiency rats increased, and the penetration enhancement effect of 2% Azone on rabbit skin of Qi deficiency was the best.
