目的:研究过氧化物酶增殖因子激活受体γ(peroxisomeproliferatorsactivatedreceptorγ,PPARγ)激活剂罗格列酮对人胃癌细胞系MKN45增殖和MMP7基因表达的影响。方法:不同浓度罗格列酮干预细胞后,用台盼蓝拒染法检测胃癌细胞株增殖活性,用RTPCR法检测MMP7基因表达情况。结果:罗格列酮作用24h明显降低MKN45细胞的存活率,与对照组比较有显著差异(P<0.05),细胞存活率随着药物浓度增加而降低;MMP7基因表达随着药物浓度增加而下降。结论:在胃癌细胞株MKN45中,罗格列酮与PPARγ作用而激活下级信号传导,抑制胃癌细胞的增殖,且下调MMP7基因的表达。
Objective: To investigate the effect of peroxisome proliferator activated receptor γ (PPARγ) activator rosiglitazone on the proliferation of human gastric cancer cell line MKN45 and the expression of MMP7 gene. Methods: After the cells were treated with different concentrations of rosiglitazone, the proliferation of gastric cancer cell lines was detected by trypan blue exclusion method. The expression of MMP7 gene was detected by RTPCR. Results: The effect of rosiglitazone on the survival rate of MKN45 cells was significantly lower than that of the control group at 24 hours (P <0.05). The cell viability decreased with the increase of the drug concentration. The expression of MMP7 decreased with the increase of the drug concentration . Conclusion: In gastric cancer cell line MKN45, rosiglitazone and PPARγ activate sub-level signaling, inhibit the proliferation of gastric cancer cells and down-regulate the expression of MMP7.
