目的 观察环氧化酶异构酶COX 1和COX 2在大鼠胃溃疡模型中的动态表达和活性变化。方法 采用乙酸胃腔内注射方法制备大鼠胃溃疡模型 ,采集溃疡诱导后 1h~ 15d的溃疡基底、边缘和正常胃组织标本 ,采用逆转录聚合酶链反应 (RT PCR)方法和Westernblot方法测定组织中COX 1和COX 2的mRNA表达及蛋白质水平 ,采用测定组织匀浆液代谢花生四烯酸产生前列腺素E的能力的方法检测标本的总COX活性。结果 溃疡诱导后溃疡基底的COX 2的mRNA和蛋白质在炎症期 (6h~ 3d)高度诱导表达并且其蛋白质在溃疡愈合期 (3~ 15d)维持较高水平。COX 1的表达变化在溃疡诱导前后相对恒定。溃疡基底组织及溃疡边缘组织匀浆液随溃疡的愈合表现出更高的总COX活性 ,而溃疡周围正常组织的COX活性变化不显著。结论 在溃疡愈合期仍保持高水平表达和活性的COX(特别是COX 2 )可能在损伤愈合和组织重建过程中起到重要作用。
Objective To observe the dynamic changes of cyclooxygenase isomerase COX 1 and COX 2 in gastric ulcer model of rats. Methods The rats model of gastric ulcer was prepared by intragastric injection of acetic acid. The base, marginal and normal gastric ulcer specimens were collected from 1 h to 15 d after induction of ulcer. The tissues were determined by reverse transcription polymerase chain reaction (RT PCR) and Western blot In COX 1 and COX 2 mRNA expression and protein levels, the determination of tissue homogenate metabolism of arachidonic acid to produce prostaglandin E method to detect the total COX activity of the sample. Results After ulcer induction, COX 2 mRNA and protein in ulcerous basal layer were highly expressed in the inflammatory phase (6h-3d) and the protein level was maintained at the healing stage of ulcer (3 ~ 15d). The change of COX 1 expression was relatively constant before and after ulcer induction. The ulcer basal tissue and ulceration marginal tissue homogenate showed higher total COX activity with the healing of ulcer, while the change of COX activity in normal tissue around the ulcer did not change significantly. Conclusions COX, especially COX 2, that maintains high levels of expression and activity during ulcer healing may play an important role in wound healing and tissue remodeling.
