应用大鼠皮肤角朊细胞的原代培养技术并结合单细胞凝胶电泳法检测氧化型染发剂的两种主要成分双氧水(H2 O2 )、对苯二胺 (PPD)及二者混合物致大鼠皮肤角朊细胞DNA的单链断裂作用。结果表明 ,染发剂及其组分均能使大鼠角朊细胞DNA发生单链断裂 ,染毒剂量与慧尾长度呈剂量 反应关系 (rH2 O2 =0 978,rPPD=0 993,rH2 O2 -PPD=0 996 ) ,说明DNA单链断裂是氧化型染发剂对皮肤细胞产生遗传毒作用的一种方式
Application of primary culture of keratinocytes in rat skin combined with single cell gel electrophoresis detection of two main components of oxidative hair dye hydrogen peroxide (H2 O2), p-phenylenediamine (PPD) and a mixture of the two caused by rats Single stranded DNA cleavage of cutaneous keratinocytes. The results showed that both the hair dye and its components could cause the single strand breaks of DNA in rat keratinocytes. The doses of the hair dye and the tail length were dose-dependent (rH2 O2 = 0 978, rPPD = 0 993, rH2 O2 -PPD = 0 996), indicating that DNA single strand breaks are a way for oxidative hair dyes to produce genotoxic effects on skin cells
