目的通过原位移植的方法制备大鼠肝癌动物模型。方法取大鼠肿瘤细胞Walker-256制备为浓度1×108个/ml,分别于Wistar大鼠肝包膜下接种0.04 ml。观察大鼠肝脏实体瘤的生长情况及肺转移的发生率。结果肝包膜下接种细胞悬液7 d可见较小肿瘤块生长;14 d开始出现肺转移,并伴有肠系膜、生殖系统的转移;21 d肺转移率达100%。肝肿瘤组织HE染色结果可见肿瘤组织呈条索及团块状排列,排列紊乱,癌细胞索之间由血窦相连,癌肿边缘的肝组织受压萎缩。肺组织切面有多个散在的圆形或卵圆形实化灶。HE染色结果可见腺体样或乳头状结构,核染色深,核仁、核膜比较清楚。
Objective To prepare rat liver cancer model by orthotopic transplantation. Methods Walker-256 tumor cells were prepared to a concentration of 1 × 108 / ml, were inoculated Wistar rat liver capsule 0.04 ml. Observe the growth of rat liver solid tumors and the incidence of lung metastases. Results Small hepatic tumor growth was observed on the 7th day after inoculation of the hepatic capsule. Pulmonary metastasis was observed on the 14th day accompanied by mesenteric and reproductive system transfer. The lung metastasis rate reached 100% on the 21st day. Hematoxylin-eosin (HE) staining showed that the tumor tissue was arranged in the shape of clumps and lumps, disordered, and connected by sinusoids. The liver tissue at the edge of the cancer swollen and contracted. Lung tissue section has a plurality of scattered round or oval-shaped stove. HE staining results can be seen glandular or papillary structure, deep nuclear staining, nucleolus, nuclear membrane more clearly.
