目的:探讨miRNA-196a-2的单核苷酸多态(single nucleotide polymorphisms,SNPs)位点rs11614913与胃癌易感性的关系。方法:采用病例-对照研究设计,研究对象为232例经组织学确诊的胃癌病例和250例按年龄、性别进行匹配的无肿瘤史的社区对照人群。采用探针法SNP基因分型技术检测miRNA-196a-2基因的C/T多态的基因型,计算rs11614913多态位点基因型在病例与对照中的分布情况,采用logistic回归计算校正相对危险度(odds ratio,OR)和95%置信区间(95%confidence interval,95%CI),评价基因型对胃癌易感性的影响。结果:miRNA-196a-2基因多态位点rs11614913 CC、CT和TT基因型在胃癌人群中的分布频率为44.0%、47.0%和9.0%,与对照组(28.8%,54.4%和16.8%)相比差异有统计学意义(均P<0.05);与CC基因型相比,携带TT或CT基因型的个体胃癌发病危险度降低(OR=0.369,95%CI:0.204~0.659;OR=0.564,95%CI:0.387~0.830);携带突变T碱基(CT/TT)的胃癌患者发生淋巴结转移的危险度降低(OR=0.618,95%CI:0.438~0.795;P=0.010)。讨论:miRNA-196a-2基因的C/T多态性与中国汉族人群胃癌的遗传易感性相关。
Objective: To investigate the relationship between single nucleotide polymorphisms (SNPs) rs11614913 and gastric cancer susceptibility in patients with miRNA-196a-2. METHODS: A case-control study was designed. The study population included 232 histologically confirmed gastric cancer cases and 250 community-free controls matched for age and sex. Genotypes of miRNA-196a-2 C / T polymorphism were detected by probe SNP genotyping, distribution of rs11614913 polymorphism loci in cases and controls was calculated, and logistic regression was used to calculate relative risk The odds ratio (OR) and 95% confidence interval (95% CI) were used to evaluate the effect of genotype on gastric cancer susceptibility. Results: The frequencies of rs11614913 genotypes of rs11614913 in miRNA-196a-2 gene loci were 44.0%, 47.0% and 9.0% in gastric cancer patients, which were significantly higher than those in control subjects (28.8%, 54.4% and 16.8% (OR = 0.369, 95% CI: 0.204-0.659, OR = 0.564, P <0.05). Compared with CC genotype, the incidence of gastric cancer with TT or CT genotype was significantly lower , 95% CI: 0.387-0.830). The risk of lymph node metastasis in patients with gastric cancer with mutation of T-base (CT / TT) was lower (OR = 0.618, 95% CI: 0.438-0.795; P = 0.010). Discussion: The C / T polymorphism of miRNA-196a-2 gene is associated with the genetic susceptibility of gastric cancer in Chinese Han population.
