目的:观察秦皮醇提物(EEFC)对D-半乳糖所致亚急性衰老小鼠的抗衰老作用。方法:将ICR小鼠60只随机分为空白组(生理盐水100 mg·kg-1),模型组(D-半乳糖100 mg·kg-1),阳性药维生素E组(D-半乳糖100 mg·kg-1+维生素E30 mg·kg-1),EEFC低剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物37.5 mg·kg-1)、中剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物75 mg·kg-1)和高剂量组(D-半乳糖100 mg·kg-1+秦皮醇提物150 mg·kg-1),每组10只。连续ig给药60 d,末次给药后摘眼球取血,之后取脑、胸腺、脾脏、肝、皮肤,测定脑、胸腺、脾脏的脏器指数;测定血清、脑、肝、皮肤超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化氢酶(CAT)、谷胱甘肽过氧化氢酶(GSH-Px)、丙二醛(MDA)、总抗氧化能力(T-AOC)。结果:模型组的脑、胸腺、脾脏指数比空白组明显降低(P<0.05,P<0.01),小鼠血清、肝脏、皮肤、脑模型组T-AOC,SOD,CAT的值与空白组明显降低,MDA升高。秦皮醇提物各剂量组与模型组相比,高剂量组的脑、胸腺、脾脏质量指数显著增加(P<0.05,P<0.01)。秦皮醇提物组血清、脑组织、肝和皮肤中的SOD,CAT活性均显著增强(P<0.05,P<0.01),MDA含量降低(P<0.05,P<0.01),T-AOC显著增强(P<0.05,P<0.01),血清和皮肤中的GSH-Px活性显著增强(P<0.05,P<0.01)。结论:秦皮醇提物具有延缓D-半乳糖所致亚急性衰老小鼠的衰老作用。
OBJECTIVE: To observe the anti-aging effects of ethanol extract of Epigenes (EEFC) on sub-acute aging mice induced by D-galactose. Methods: Sixty ICR mice were randomly divided into blank group (100 mg · kg-1 normal saline), model group (D-galactose 100 mg · kg -1) and vitamin E group (D-galactose 100 low-dose EEFC group (100 mg · kg-1 of D-galactose and 37.5 mg · kg-1 of hydrocortisone extract), medium dose group (D-half Lactose 100 mg · kg-1 + ethanol extract 75 mg · kg-1) and high-dose group (D-galactose 100 mg · kg-1 + hydrocortisone 150 mg · kg-1) only. The rats were sacrificed for 60 d after the last administration, and the eyeballs were taken for blood after the last administration. The brain, thymus, spleen, liver and skin were taken for determination of the organ index of the brain, thymus and spleen. The serum, brain, liver and skin superoxide (SOD, CAT, CAT, GSH-Px, MDA, total antioxidant capacity (T- AOC). Results: The index of brain, thymus and spleen in the model group was significantly lower than that in the blank group (P <0.05, P <0.01). The levels of T-AOC, SOD and CAT in the serum, liver, Reduce, MDA increased. Compared with model group, the contents of brain, thymus and spleen in high dose group were significantly increased (P <0.05, P <0.01). The activities of SOD and CAT in serum, brain tissue, liver and skin were significantly increased (P <0.05, P <0.01), the contents of MDA decreased (P <0.05, P <0.01) (P <0.05, P <0.01). GSH-Px activity in serum and skin was significantly increased (P <0.05, P <0.01). Conclusion: Qinpi alcohol extract can delay the aging effect of sub-acute aging mice induced by D-galactose.
