为分离并鉴定中国大陆输入性B3基因型麻疹病毒。使用淋巴信号激活因子转染的非洲绿猴肾细胞(Vero/Slam细胞)对该麻疹病例的咽拭子标本进行病毒分离,经逆转录聚合酶链反应(Reverse transcript polymerase chain reaction,RT-PCR)方法扩增出核蛋白(Nucleoprotein,N)羧基(COOH)端核苷酸片段,并进行序列测定和分析。结果表明这两株B3基因型麻疹病毒分离株和世界卫生组织(World Health Organization,WHO)B3基因型参考株MVi/Ibadan.NGA/97在基因亲缘性关系树上同属一个分支,核苷酸同源性均为98%;与麻疹核苷酸序列数据管理系统(Measles nucleotide surveillance,MeaNS)中的病毒序列进行比对,结果表明本研究分离的两株麻疹病毒与2013年同期在澳大利亚、日本、韩国,中国香港地区,菲律宾、伊朗等国家流行的B3基因型麻疹病毒亲缘性关系为100%。在中国上海分离到的麻疹毒株为B3基因型。是中国自1993年开展病毒学监测以来首次分离到B3基因型病毒,为国外输入性的麻疹病毒。本研究中分离到的B3基因型麻疹病毒对于积累中国麻疹分子流行病学基线数据具有很重要的意义,也有助于全国及全球麻疹病毒的传播及溯源。
In order to isolate and identify the imported genotype B3 measles virus in mainland China. The results showed that the two genotype B3 measles virus isolates and the World Health Organization (WHO) B3 genotype reference strain MVi / Ibadan.GA / 97 belong to the same gene relationship tree, and the nucleotides are the same The results showed that the two measles viruses isolated in this study were identical with those in the same period in 2013 in Australia, Japan, Japan, Korea, China Hong Kong, the Philippines, Iran and other countries the prevalence of B3 genotype measles virus was 100%. The measles isolate isolated in Shanghai, China, is the B3 genotype. It is the first time that China has isolated B3 genotype virus since its virological surveillance in 1993 and is a foreign measles virus imported from abroad. The B3 genotype measles virus isolated in this study is of great significance for the accumulation of molecular epidemiological baseline data of measles in China and also contributes to the spread and traceability of measles virus nationwide and globally.
