目的观察复方菖蒲益智汤对脑缺血再灌注损伤小鼠急性期脑组织谷氨酸及血清钙调神经磷酸酶(CaN)活性的影响。方法将实验小鼠随机分为假手术组、模型组、复方菖蒲益智汤高剂量组(高剂量组)、复方菖蒲益智汤低剂量组(低剂量组)、尼莫地平对照组(尼莫地平组),每组10只。采用双侧颈总动脉夹闭的方法制备脑缺血再灌注损伤小鼠模型,给药14天后,观察海马区脑组织的病理形态学变化,测定脑组织谷氨酸及血清CaN(钙调神经磷酸酶)活性。结果各用药组小鼠海马区锥体细胞病变较模型组均有所改善,高剂量组改善最为显著;高剂量组小鼠脑组织谷氨酸活性、血清CaN活性均低于模型组、低剂量组及尼莫地平组(P<0.05)。结论复方菖蒲益智汤能够降低脑缺血再灌注小鼠脑组织谷氨酸、血清CaN活性,对于小鼠脑缺血再灌注损伤有保护作用。
Objective To observe the effect of compound aconite and Yizhi decoction on glutamate and serum calcineurin (CaN) activity in brain of acute cerebral ischemia-reperfusion mice. Methods The experimental mice were randomly divided into sham operation group, model group, high-dose compound calabash Yizhi decoction group (high-dose group), low-dose compound group Changyu Decoction (low dose group), nimodipine control group Mupirin group), each group of 10. The model of cerebral ischemia-reperfusion injury was established by bilateral common carotid artery occlusion. After 14 days of administration, pathological changes of hippocampus were observed. Glutamate and serum CaN Phosphatase) activity. Results The pathological changes of pyramidal cells in hippocampus of mice in each treatment group were improved compared with those in model group and the improvement in high dose group was the most significant. The glutamate activity and serum CaN activity in high dose group were lower than those in model group and low dose Group and nimodipine group (P <0.05). Conclusion Compound Prescription Guichang Decoction can reduce the activity of glutamate and serum CaN in the brain tissue of mice with cerebral ischemia and reperfusion, and has the protective effect on the cerebral ischemia-reperfusion injury in mice.
