目的:观察细梗胡枝子总黄酮对急性应激血瘀大鼠血液流变学和血液凝固的影响。方法:利用尾静脉注射阿霉素复制大鼠微小病变肾病模型,采用在2次皮下注射肾上腺素1 mg.kg-1之间将大鼠置于冰水中5 min的方法建立急性应激血瘀大鼠模型。观察胡枝子总黄酮对模型大鼠全血黏度(TBAV)、全血还原黏度(TBRV)、红细胞压积(Hct)、红细胞聚集性(EAI)等血液流变学指标的作用,同时测定凝血时间(CT)、凝血酶原时间(PT)、活化部分凝血激酶时间(APTT)、凝血酶时间(TT)及血小板聚集率和血小板解聚时间。结果:胡枝子总黄酮5.5,2.25 g.kg-1能明显降低高、中、低切变率下全血黏度,降低中、低切变率下全血还原黏度和红细胞压积,降低红细胞聚集性(与模型组比较P<0.05或P<0.01)。5.5,2.25 mg.kg-1可使模型大鼠CT,PT,APTT,TT明显延长(与模型组比较P<0.01),并使血小板最大聚集率降低和血小板开始解聚的时间缩短(P<0.01),且作用呈剂量依赖性。结论:胡枝子总黄酮对血液流变学指标和血液凝固有明显的影响,具有活血化瘀作用。
OBJECTIVE: To observe the effect of total flavonoids of Lespedeza brassicae on blood rheology and blood coagulation in rats with acute stress blood stasis. METHODS: Rat model of minimal change nephropathy was induced by tail vein injection of adriamycin. Acute stress was established by placing rats in ice water for 5 min between two subcutaneous injections of epinephrine 1 mg.kg-1. Rat model. To observe the effects of total flavonoids of Lespedeza zhizhi on hemorheological indexes such as whole blood viscosity (TBAV), whole blood reduced viscosity (TBRV), hematocrit (Hct), and red blood cell aggregation (EAI) in model rats and to determine the clotting time (CT), prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT) and platelet aggregation rate and platelet depolymerization time. RESULTS: The total flavonoids of L. chinensis 5.5, 2.25 g.kg-1 significantly reduced the viscosity of whole blood at high, middle and low shear rates, reduced the reduced viscosity and hematocrit of whole blood at medium and low shear rates, and reduced red blood cell aggregation. Sex (P<0.05 or P<0.01 compared with the model group). 5.5, 2.25 mg.kg-1 significantly prolonged the CT, PT, APTT, and TT in the model rats (P<0.01 compared with the model group), and reduced the maximum platelet aggregation rate and the time for platelet depolymerization (P< 0.01), and the effect was dose-dependent. Conclusion: The total flavonoids of L. chinensis have obvious effects on hemorheology indexes and blood coagulation, and it has the effect of promoting blood circulation and removing blood stasis.
