Purpose: To investigate the results of the interferon-gamma(IFN-γ) gene transfer andtransient expression in human Tenon’s capsule fibroblast in vitro in order to find a way togene therapy in vivo. Method: Using LipofectAMINE, IFN-γ gene was transferred inhuman Tenon’s capsule fibroblasts with plasmid pcDNAS IFN-γ. Its mRNA transcriptionand protein expression were determined by RT-PCR and flow cytometry assayrespectively.Result: The human Tenon’s capsule fibroblasts transferred the IFN-γ gene can expressthe IFN-γ in transcription and protein level transiently.Conclusion: IFN-γ gene can be transferred successfully and expressed efficiently inhuman tenon’s capsule fibroblast in vitro.
