目的:研究乏氧诱导因子-1α(HIF-1α)基因及糖酵解相关基因己糖激酶-Ⅱ(HK-Ⅱ)、乳酸脱氢酶-Ⅴ(LDH-Ⅴ)在原发性肝细胞癌(HCC)组织中的表达状态,并阐释三者对HCC生物学行为的影响。方法:收集行手术治疗的56例新鲜肝癌组织标本,通过免疫组织化学技术检测HIF-1α、HK-Ⅱ和LDH-Ⅴ等基因在肝癌组织中的蛋白表达情况,并分析其与HCC临床病理学指标之间的关系。结果:在肝细胞癌组织中HIF-1α、HK-Ⅱ和LDH-Ⅴ蛋白的阳性表达率分别为71.4%、75.0%和67.9%。HIF-1α、HK-Ⅱ及LDH-Ⅴ三者的阳性表达均与HCC的有无肝硬化背景、肿瘤细胞的分化程度、临床分期、有无肝门淋巴结转移、有无门脉/胆道癌栓的形成、有无静脉侵袭及包膜完整与否等因素相关,P<0.05;而与HCC患者的性别、年龄、HBsAg阳性或阴性、血清AFP高低及癌灶的直径大小等指标无关,P>0.05。结论:HK-Ⅱ、LDH-Ⅴ基因与HIF-1α基因的同步表达在HCC的发生、发展中可能发挥协调作用,共同促进肝癌细胞的侵袭与转移,反映了HCC的生物学行为,可以将三者作为预测HCC预后的重要生物学指标。
Objective: To investigate the effect of hypoxia-inducible factor-1α (HIF-1α) gene and glycoprotein related gene hexokinase-Ⅱ (HK-Ⅱ) and lactate dehydrogenase-V (LDH-Ⅴ) on primary hepatocellular carcinoma (HCC) tissues and to explain the impact of the three on the biological behavior of HCC. Methods: Fifty-six fresh HCC tissues were collected and the protein expression of HIF-1α, HK-Ⅱ and LDH-Ⅴ were detected by immunohistochemistry in HCC tissues. The correlations between HIF-1α, The relationship between the indicators. Results: The positive rates of HIF-1α, HK-Ⅱ and LDH-Ⅴ proteins in HCC tissues were 71.4%, 75.0% and 67.9%, respectively. The positive expression of HIF-1α, HK-Ⅱ and LDH-V were correlated with the liver cirrhosis background, tumor cell differentiation, clinical stage, hilar lymph node metastasis, portal vein / cholangiocarcinoma (P <0.05), but not related to the gender, age, HBsAg positive or negative, serum AFP level and the size of the foci of HCC (P> 0.05), P> 0.05. Conclusion: The simultaneous expression of HK-Ⅱ, LDH-Ⅴ gene and HIF-1α gene may play a coordinating role in the occurrence and development of HCC, which together promote the invasion and metastasis of HCC. It reflects the biological behavior of HCC, As a predictor of prognosis of HCC important biological indicators.
