采用给禁食大鼠脑室灌注溶于不同溶剂中的GLP-1(7-36)的方法,观察GLP-1在中枢对大鼠血糖及血糖调节激素浓度的影响。结果表明:脑室灌注溶于生理盐水的GLP-1与单纯灌注生理盐水相比,血糖无明显变化,但1小时后,血中胰岛素、胰升糖素浓度均明显降低;脑室灌注溶于30%葡萄糖的GLP-1与单纯灌注30%葡萄糖相比,血糖明显升高,在30’、45’、60’有显著性差异,血中胰岛素浓度明显低于对照组,胰升糖素浓度无变化。这些实验说明GLP-1参与了中枢神经系统对胰岛分泌激素功能的调节。
The effect of GLP-1 on the blood glucose and glucose-regulated hormones in rats was observed by perfusing GLP-1 (7-36) dissolved in different solvents into the ventricles of rats. The results showed that GLP-1 dissolved in physiological saline had no significant change in blood glucose compared with normal saline, but the concentration of insulin and glucagon in blood decreased significantly after 1 hour. Glucose GLP-1 compared with the simple perfusion of 30% glucose, blood glucose was significantly increased at 30 ’, 45’, 60 ’have significant differences in blood insulin levels were significantly lower than the control group, no change in glucagon concentration . These experiments demonstrate that GLP-1 is involved in the regulation of insulin secretion hormone function by the central nervous system.
