目的:探讨左归丸对妊娠糖尿病大鼠胎盘组织11β-羟基类固醇脱氢酶1(11β-HSD1)基因表达水平的影响。方法:育龄雌性SD大鼠受孕后随机分为正常妊娠组(C)、妊娠糖尿组(GDM)、胰岛素干预组(INS)、左归丸干预组(ZGW)、左归丸联合胰岛素干预组(F),每组10只;受孕当日GDM组和3个干预组按45 mg·kg-1ip链脲佐菌素(STZ)复制妊娠糖尿病模型。并于孕4 d开始给药干预(左归丸10 g·kg-1ig,胰岛素20 U·kg-1ih)。孕19 d断头取血,葡萄糖氧化酶法测定血糖;酶联免疫吸附法检测血清皮质酮、胰岛素;采用RT-q PCR技术检测胎盘11β-HSD1基因mRNA的表达水平,并进行定量分析。结果:与正常妊娠组比较,GDM组血糖水平、血清胰岛素浓度、皮质酮水平、胎盘11β-HSD1基因mRNA的表达水平均明显升高,差异有统计学意义(P<0.01);与妊娠糖尿组比较,左归丸干预组血糖水平、血清胰岛素浓度、皮质酮水平、胎盘11β-HSD1基因mRNA的表达水平均明显降低(P<0.05,P<0.01),但仍高于正常妊娠组,联合组治疗效果优于左归丸组。结论:左归丸可降低妊娠期糖尿病大鼠血糖水平,胎盘部位有11β-HSD1基因的表达,妊娠糖尿病病理状态可影响11β-HSD1基因在胎盘的表达水平,左归丸有下调11β-HSD1基因表达水平的作用,推测可能是其改善GDM大鼠胰岛素抵抗的机制之一。
Objective: To investigate the effects of Zuo Gui Pill on 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1) gene expression in placenta of gestational diabetes mellitus (DM). Methods: Pregnant female SD rats were randomly divided into normal pregnancy group (C), gestational diabetes mellitus group (GDM), insulin intervention group (ZGW), Zuogui pill combined with insulin intervention group F) with 10 mice in each group. The gestational diabetes mellitus model was induced by 45 mg · kg-1 streptozotocin (STZ) in GDM group and 3 intervention groups on the day of conception. The intervention was started on the 4th day of pregnancy (Zuogui pill 10 g · kg-1ig, insulin 20 U · kg-1ih). Serum corticosterone and insulin were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA expression level of 11β-HSD1 in placenta was detected by RT-q PCR and quantified. Results: Compared with the normal pregnancy group, the levels of blood glucose, serum insulin, corticosterone and placental 11β-HSD1 mRNA in GDM group were significantly increased (P <0.01) Compared with the normal pregnancy group and the combination group, the blood glucose level, serum insulin level and corticosterone level in the Zuoguiwan intervention group were significantly decreased (P <0.05, P <0.01) The treatment effect is better than Zuo Gui Wan group. Conclusion: Zuogui pill can reduce the level of blood glucose in gestational diabetes mellitus, the expression of 11β-HSD1 gene in placenta. The gestational diabetes mellitus can affect the expression level of 11β-HSD1 gene in the placenta. Zuogui pill down-regulated 11β-HSD1 gene Expression of the role, presumed that it may be one of the mechanisms to improve insulin resistance in GDM rats.
