以18个甜橙品种为材料,采用克隆测序法对其核糖体DNA ITS进行序列测定,并用DNA Star软件进行序列分析.结果显示,甜橙ITS序列的长度为693~694bp,其中ITS1长272bp,5.8S长度为164bp,ITS2长度为257~258bp.甜橙品种间ITS序列的相似性较高,相似率为97.7%~100%.同时发现18个甜橙品种ITS序列中存在20个碱基变异位点,其中ITS1和ITS2序列中各含10个,而5.8S序列中没有碱基变异.根据这些碱基变异位点可鉴别出供试材料中的12个甜橙品种.
Totally 18 sweet orange cultivars were used as materials to sequence the ITS of ribosomal DNA by cloning and sequencing and sequenced with DNA Star software.The results showed that the length of ITS sequence of sweet orange was 693-694 bp, 5.8S length was 164bp and ITS2 length was 257 ~ 258bp. The similarity of ITS sequences between sweet orange cultivars was high, and the similarity rate was 97.7% -100% .There were 20 base variations in the ITS sequences of 18 sweet orange cultivars Sites, including ITS1 and ITS2 sequences containing 10, respectively, and 5.8S sequences without base variation.According to these base variation sites can be identified in the test material 12 sweet orange varieties.
