根据蜻蜓凤梨转录组信息,通过RACE技术(rapid amplification of cDNA ends,RACE)获得蜻蜓凤梨中一个DELLA蛋白的cDNA序列,并将其命名为AfGAI。AfGAI cDNA全长1 960 bp,开放阅读框为1 764 bp,编码587个氨基酸。氨基酸序列分析结果显示,AfGAI蛋白含有DELLA蛋白家族共有的功能结构域:DELLA结构域和GRAS结构域,与菠萝(Ananas comosus)、海枣(Phoenix dactylifera)、油棕(Elaeis guineensis)、巨桉(Eucalyptus grandis)、陆地棉(Gossypium hirsutum)中的GAI同源性分别为90%、73%、73%、65%、59%。进一步构建了35S:AfGAI过表达载体,为解析凤梨科植物开花机理提供理论依据。
According to the dragonfly pineapple transcriptome information, a cDNA sequence of a DELLA protein from the dragonfly pineapple was obtained by RACE (Rapid Amplification of cDNA ends, RACE) and named it AfGAI. The cDNA of AfGAI is 1 960 bp in length with a 1 764 bp open reading frame encoding 587 amino acids. The results of amino acid sequence analysis showed that AfGAI protein shared the functional domains of DELLA protein family: DELLA domain and GRAS domain, which were closely related to Ananas comosus, Phoenix dactylifera, Elaeis guineensis, Eucalyptus grandis and Gossypium hirsutum were 90%, 73%, 73%, 65% and 59%, respectively. 35S: AfGAI overexpression vector was further constructed to provide a theoretical basis for analyzing the flowering mechanism of bromeliads.
