观察雌激素对卵巢癌细胞的增殖调节。利用卵巢上皮性癌AO、3AO细胞,采用增殖细胞核抗原免疫组化检测法及3H-胸腺嘧啶掺入率测定法观察了不同浓度的雌二醇(E2)对卵巢癌细胞增殖的影响。结果:两株细胞对10-5~1013 mol/L之间的17-β-E2具有相似的剂量反应。AO细胞在10-7~10-9mol/L浓度范围中增殖明显,而超出该范围这种作用由促进转向抑制; 3AO细胞在 10-9~ 10-13 mol/ L之间增殖明显。两者在10-5mol/ L的 E2的作用下均呈现明显的抑制作用、结论:雌激素对卵巢癌的增殖作用具有剂量依赖性,高浓度的雌激素对癌细胞有明显的抑制作用。
To observe the estrogen on ovarian cancer cell proliferation regulation. The effect of different concentrations of estradiol (E2) on the proliferation of ovarian cancer cells was observed by immunohistochemistry of proliferating cell nuclear antigen and 3H-thymidine incorporation assay using ovarian epithelial carcinoma AO and 3AO cells. RESULTS: The two cells had a similar dose response to 17-β-E2 between 10-5 and 1013 mol / L. AO cells proliferation significantly in the concentration range of 10-7 ~ 10-9mol / L, and beyond this range by promoting the steering inhibition; 3AO cells in 10-9 ~ 10-13 mol / L proliferation significantly. Both of them showed obvious inhibitory effect under the effect of E2 of 10-5mol / L. Conclusion: Estrogen has a dose-dependent effect on the proliferation of ovarian cancer. High concentrations of estrogen have obvious inhibitory effect on cancer cells.
