目的:获得一种高效简易的微生物辅助脱去瓜蒌瓤的方法,并对其条件进行优化。方法:分别利用酶解-蒽酮比色法、苯酚-硫酸法、DNS法及双缩脲比色法对瓜蒌瓤中成分进行分析测定,确定微生物的选择。利用正交试验设计法,将菌液浓度、菌液体积、样品质量以及静置时间等4个因子按照正交表安排试验,最终对结果进行计算分析。结果:瓜蒌瓤中淀粉含量为36.08%,总糖含量为43.75%,还原性糖含量为27.68%,粗蛋白含量为21%。选用安琪酵母菌为试验辅助菌,在菌液浓度为2.01×107 cuf/m L、菌液体积为70 m L、样品质量为50 g、静置时间为96 h时,洗脱效果达到最优,洗脱率达到98%。
OBJECTIVE: To obtain an efficient and simple method for microbial removal of melon and its conditions. Methods: The components of melon were determined by enzymatic hydrolysis - anthrone colorimetric method, phenol - sulfuric acid method, DNS method and biuret colorimetric method, respectively, to determine the choice of microorganisms. Using orthogonal test design method, the four factors of bacterial liquid concentration, bacterial liquid volume, sample quality and standing time were arranged according to the orthogonal test table, and finally the results were calculated and analyzed. Results: Starch content in melon was 36.08%, total sugar content was 43.75%, reducing sugar content was 27.68% and crude protein content was 21%. Antagonistic yeast was selected as the auxiliary test bacteria. When the concentration of bacteria was 2.01 × 107 cuf / mL, the volume of bacterial liquid was 70 mL, the sample mass was 50 g, and the standing time was 96 h, the elution effect reached the maximum Excellent, elution rate of 98%.
