目的:探讨不同植物激素配比、Vc对薄荷茎段不定芽诱导的影响,及不同浓度NaN_3对其愈伤组织的诱变效果,为薄荷离体化学诱变体系的建立奠定一定的理论研究基础。方法:以薄荷试管苗节间部分为试验材料,研究不同浓度的TDZ、6-BA、NAA及Vc对薄荷不定芽诱导的影响,在筛选出最佳诱导配方的基础上,用不同浓度的NaN_3(0、2、4、6、8、10、12、14和16 mg/L)对薄荷的愈伤组织进行诱变处理。结果:培养基MS+0.1 mg/L TDZ+0.2mg/L NAA+1 mg/L Vc+30 g/L蔗糖+5.5 g/L琼脂诱导愈伤组织和不定芽分化的效果最佳,NaN_3诱导薄荷愈伤组织的LD50为14 mg/L处理10 d,12 mg/L处理20 d,10 mg/L处理30 d,诱变后愈伤组织经分化形成完整植株,通过对比再生植株形态学差异,筛选出变异植株81株。结论:初步建立了薄荷的NaN_3离体化学诱变体系。
OBJECTIVE: To investigate the effects of different plant hormones, Vc on the induction of adventitious buds of peppermint stems, and the mutagenic effects of different concentrations of NaN_3 on callus, and lay a theoretical foundation for the establishment of the chemical mutagenesis system in vitro . Methods: The effects of different concentrations of TDZ, 6-BA, NAA and Vc on the induction of adventitious buds of peppermint were studied in the internodes of peppermint plantlets. On the basis of selecting the best inducing formula and using different concentrations of NaN3 (0, 2, 4, 6, 8, 10, 12, 14 and 16 mg / L) were used to mutagenize mint callus. Results: The best medium for inducing differentiation of callus and adventitious buds was MS + 0.1 mg / L TDZ + 0.2 mg / L NAA + 1 mg / L Vc + 30 g / L sucrose + 5.5 g / L agar. NaN 3 induction The callus of M. mongolica had the LD50 of 14 mg / L for 10 days, 12 mg / L for 20 days and 10 mg / L for 30 days. After callus induction, the callus was differentiated to form a complete plant. , 81 mutant plants were screened out. Conclusion: The chemical modification of NaN_3 in vitro has been preliminarily established.
