c-Myc和Bin1在人食管鳞状细胞癌组织中的表达及其临床意义

目的:研究人食管鳞状细胞癌(esophageal squamous cell cancer,ESCC)组织及其癌旁组织中c-Myc和Bin1基因的表达情况,并分析其临床意义。方法:收集2013年4月至2014年3月在河北医科大学第四医院胸外科行肿瘤切除并经病理证实的ESCC患者肿瘤组织和相应癌旁组织标本各54例,使用RT-PCR和免疫组织化学法检测ESCC组织和癌旁组织中c-Myc和Bin1基因的mRNA和蛋白的表达。结果:与癌旁组织相比,ESCC组织中的c-Myc mRNA表达水平和蛋白阳性表达率均显著升高[(0.34±0.29)vs(0.17±0.16),P<0.001;55.56%vs 33.33%,P=0.033],Bin1 mRNA表达水平和蛋白阳性表达率均显著降低[(0.25±0.19)vs(0.33±0.20),P<0.001;57.41%vs 84.48%,P=0.007]。ESCC组织中c-Myc mRNA的表达与Bin1 mRNA的表达呈显著负相关关系(r=0.790,P<0.001),c-Myc蛋白表达与Bin1蛋白表达也呈显著负相关关系(P=0.019)。c-Myc和Bin1蛋白表达均与患者TNM分期、肿瘤侵犯深度、分化程度、淋巴结转移相关。结论:人ESCC患者肿瘤组织中c-Myc呈高表达,而Bin1呈低表达,两者表达呈负相关,均与ESCC进展和淋巴结转移有关联。 Objective: To study the expression of c-Myc and Bin1 in human esophageal squamous cell carcinoma (ESCC) tissues and their adjacent tissues, and to analyze the clinical significance. Methods: From April 2013 to March 2014, 54 cases of tumor tissues and corresponding paracancerous tissues in ESCC patients who underwent tumor resection and pathology in the thoracic surgery department of the Fourth Hospital of Hebei Medical University from April 2013 to March 2014 were collected. RT-PCR and immunohistochemistry The mRNA and protein expressions of c-Myc and Bin1 in ESCC tissues and adjacent tissues were detected by chemical methods. Results: The expression of c-Myc mRNA and protein in esophageal squamous cell carcinoma were significantly higher than those in paracancerous tissues [(0.34 ± 0.29) vs (0.17 ± 0.16), P <0.001; 55.56% vs 33.33% , P = 0.033]. The expression of Bin1 mRNA and the positive rate of protein expression were significantly lower than those in the control group [(0.25 ± 0.19) vs (0.33 ± 0.20), P <0.001; 57.41% vs 84.48%, P = 0.007]. There was a significant negative correlation between c-Myc mRNA expression and Bin1 mRNA expression in ESCC (r = 0.790, P <0.001), and negative correlation between c-Myc protein expression and Bin1 protein expression (P = 0.019). The expressions of c-Myc and Bin1 were correlated with TNM stage, tumor invasion depth, differentiation and lymph node metastasis. Conclusions: The expression of c-Myc in human ESCC is high, while that in Bin1 is low. The expression of c-Myc is negatively correlated with the progress of ESCC and lymph node metastasis.
