目的:探讨葛根素对糖尿病肾病(DN)大鼠肾病变的保护作用及其机制。方法:以高脂饲料喂养联合低剂量链脲佐菌素(STZ)复制DN大鼠模型,动物分为正常对照组、模型组、3个葛根素治疗组、依那普利治疗组、消渴丸治疗组。8周后检测空腹血糖、尿素氮、血肌酐(SCr)、微量白蛋白/尿肌酐比值(U.A/C)和总蛋白/尿肌酐比值(UTP/C)等;Masson染色法观察肾组织形态学改变;免疫组织化学检测肾组织纤维连接蛋白(FN)与IV胶原蛋白(IV-C)的表达。结果:ig给药,葛根素可改善DN大鼠肾脏功能和肾组织形态学异常,降低肾组织中FN与IV-C的表达。结论:葛根素对DN大鼠肾脏具有保护作用,此作用可能与下调肾脏组织FN与IV-C的表达有关。
Objective: To investigate the protective effect of puerarin on nephropathy in diabetic nephropathy (DN) rats and its mechanism. Methods: The DN rats were induced by high-fat diet combined with low-dose streptozotocin (STZ). The animals were divided into normal control group, model group, three puerarin-treated groups, enalapril-treated group, Pill treatment group. After 8 weeks, fasting blood glucose, urea nitrogen, serum creatinine (SCr), microalbuminuria / urine creatinine ratio (UA / C) and total protein / urine creatinine ratio (UTP / C) were measured. Masson staining was used to observe the morphological changes of kidney The expressions of FN and IV-C in renal tissue were detected by immunohistochemistry. Results: ig administration, puerarin can improve renal function and renal morphological abnormalities in DN rats, and reduce the expression of FN and IV-C in renal tissues. CONCLUSION: Puerarin has a protective effect on the kidneys of DN rats, which may be related to the down-regulation of FN and IV-C expression in kidney.
