目的了解生后不同时期BALB/c小鼠淋巴结内促甲状腺激素受体(TSHR)的表达状况及给予新生期BALB/c小鼠己烯雌酚(DES)后对该表达所产生的影响,并借此探讨生后不同时期小鼠淋巴结内促甲状腺激素(TSH)的可能来源及DES对TSH表达所产生影响的可能机制。方法选取同窝及体重差异小于5%的新生BALB/c小鼠,随机分为DES组与对照组,雌、雄各选30对,即共取小鼠120只,在生后24h内,DES组小鼠颈背部皮下注射DES,每隔24h注射1次,共连续注射5次;对照组小鼠平行注射等量无菌豆油。分别在生后第7、14、21、35、49和第63天将小鼠处死,每个时间点雌、雄各取5对小鼠,取其颈深淋巴结进行常规石蜡包埋、切片及TSHR的免疫组织化学检测。结果无论是对照组还是DES组,小鼠从生后第7天到第63天,淋巴结内均可见到TSHR阳性细胞,阳性表达在雌性和雄性小鼠分别于生后第49天和第35天达到峰值。同一时间点对照组和DES组相比较发现,无论是平均吸光度(AA)还是积分吸光度(IA),DES组均高于对照组。在雌性小鼠,两组间AA和IA在生后各时间点均有统计学差异;在雄性小鼠,两组间AA在生后第14、21、35和第49天有统计学差异,IA在第14、21、35、49和第63天有统计学差异。结论 BALB/c小鼠淋巴结细胞在生后不同时期均具有结合垂体前叶来源TSH的受体基础。新生期接触DES可导致小鼠淋巴结内TSHR阳性表达增加,雌性小鼠大于雄性小鼠。而DES对雌性和雄性小鼠淋巴结内TSHR表达的影响均可持续至小鼠成年期。
Objective To investigate the expression of thyroid-stimulating hormone receptor (TSHR) in BALB / c mice lymph nodes and the effect of administering diethylstilbestrol (DES) in neonatal BALB / c mice at different stages of postnatal life. The Possible Sources of Thyroid Hormone (TSH) in Mouse Lymph Nodes at Different Stages and the Possible Mechanism of DES on TSH Expression. Methods Neonatal BALB / c mice with the same litter size and body weight less than 5% were randomly divided into DES group and control group, with 30 pairs of female and male, 120 mice were selected. Within 24 hours after birth, DES The mice were injected subcutaneously with DES on the back of the neck and injected once every 24 hours for 5 consecutive injections. The mice in the control group were injected with the same amount of sterile soybean oil in parallel. Mice were sacrificed on the 7th, 14th, 21st, 35th, 49th and 63rd days after birth, respectively. Five male and female mice were sacrificed at each time point. The deep cervical lymph nodes were paraffin embedded, Immunohistochemical detection of TSHR. Results TSHR positive cells were found in the lymph nodes from the 7th day to the 63rd day after birth in both control and DES groups. The positive expression of TSHR was observed in the female and male mice on day 49 and day 35 Peak. Compared with the DES group, the mean absorbance (AA) and integral absorbance (IA) of the control group and the DES group were higher than those of the control group at the same time points. In female mice, AA and IA were statistically different between the two groups at different time points; in male mice, there was a significant difference in AA between the two groups on the 14th, 21st, 35th and 49th day after birth IA had statistical difference on the 14th, 21st, 35th, 49th and 63rd days. Conclusion The lymph node cells of BALB / c mice have the receptor basis of TSH derived from the anterior pituitary at different stages after birth. Neonatal exposure to DES results in an increase in TSHR expression in lymph nodes of mice, which is greater in female mice than in male mice. However, the effect of DES on the expression of TSHR in the lymph nodes of both female and male mice can be continued until the adult mice.
