目的 建立一种经圆窗慢性电极观察纪录听神经自发性电活动的方法 ,并观察水杨酸钠致听神经自发性电活动增加效应的变化规律以及耳鸣治疗药物对这种变化的影响。方法 选用体质量 30 0~ 45 0 g花色豚鼠 ,预先埋置左圆窗电极 ,以电生理仪记录圆窗电极的自发性电活动变化 ,计算平均电压值(MIV)及C1~ 4共 4个频段的功率谱值 (MPBV )。观察豚鼠注射水杨酸钠 ( 4 0 0mg/kg)后AP阈值、MIV、C1~ 4MPBV随时间的变化规律以及各耳鸣治疗药物对已发生变化的AP阈值、MIV和C1MPBV的影响。结果 利用圆窗电极可以较好的观察到听神经自发性电活动。注射水杨酸钠后AP阈值在 3h后达到最高 ,2 4h后即已恢复正常。左耳圆窗区自发性电活动的MIV及C1频谱区的MPBV也在注射水杨酸钠 3h后增加到最大 ,但需 48h后才恢复到水杨酸钠注射前的水平。C2~ 4频段的功率谱在注射水杨酸钠前后无明显变化。注射所有耳鸣治疗药物前后AP阈值变化均不明显 ,注射利多卡因和盐酸氟桂嗪前后MIV和C1MPBV差异显著 ,注射生理盐水和尼莫地平前后则无明显变化。结论 经圆窗电极可以较好的纪录到豚鼠听神经自发性电活动的情况 ,水杨酸钠可以导致豚鼠圆窗区自发性电活动强度明显增加 ,这种效应可被利多卡因、盐酸氟桂嗪阻断 ,而尼莫地平和生理盐水不
Objective To establish a method of recording the spontaneous electrical activity of the auditory nerve via the round window chronic electrode and to observe the changes of spontaneous electrical activity induced by sodium salicylate and the effect of tinnitus treatment on this change. Methods The guinea pigs with the body weight of 30 ~ 45 g were used to embed the left circular window electrodes. The spontaneous electrical activities of the round window electrodes were recorded by electrophysiological method. The average voltage (MIV) and the total number of C1 ~ 4 Frequency band power spectrum (MPBV). The changes of AP threshold, MIV and C1 ~ 4 MPBV in guinea pigs after sodium salicylate injection (400 mg / kg) were observed, and the effect of each tinnitus treatment on AP threshold, MIV and C1 MPBV changed. Results The use of circular window electrode can be better observed spontaneous auditory nerve activity. AP threshold after sodium salicylate injection reached the highest after 3h, returned to normal after 24h. MPV of MIV and C1 in the left ear window window increased to the maximum at 3h after injection of sodium salicylate, but returned to the level before sodium salicylate injection 48h later. The power spectrum of C2 ~ 4 bands did not change significantly before and after injection of sodium salicylate. Before and after injection of all tinnitus drugs AP threshold changes were not obvious, injection of lidocaine and flunarizine hydrochloride before and after MIV and C1MPBV significant difference, before and after injection of saline and nimodipine no significant change. Conclusions The spontaneous electrical activity of the auditory nerve of guinea pigs can be well documented by the round window electrodes. Sodium salicylate can significantly increase the spontaneous electrical activity in the window area of guinea pigs. This effect can be blocked by lidocaine, Ozine blocks, while nimodipine and saline do not
