研究猪带绦虫重组BCG-TSOL18疫苗免疫仔猪,猪带绦虫卵攻击后诱导其保护性免疫应答的情况。将12头40d龄健康仔猪均分为3组:分别为1011 CFU重组BCG-TSOL18疫苗灌胃组、BCG对照组和PBS对照组。每2周免疫1次,共免疫2次。于末次免疫4周后用猪带绦虫卵进行攻击感染,3月后剖杀仔猪,分离囊尾蚴,计算减蚴率;取前腔静脉血,分离血清和制备外周血单个核细胞(peripheral blood monouclear cell,PBMC)。采用ELISA法检测血清IgG、IgG1及IgG2a水平以及PBMC培养上清液中IL-2、IFN-γ、IL-4及IL-10的水平;MTT比色法检测PBMC增殖水平。重组BCG-TSOL18疫苗免疫组猪带绦虫卵攻击后诱导仔猪获得79.44%的保护力。与BCG和PBS对照组比较,重组BCG-TSOL18疫苗组血清IgG、IgG2a水平显著升高(t=13.026,P=0.000;t=13.253,P=0.000;t=7.284,P=0.000;t=8.453,P=0.000),IgG1水平显著降低(t=-9.118,P=0.000;t=-10.372,P=0.000);抗原刺激下PBMC显著增殖(t=14.929,P=0.000;t=14.665,P=0.000);PBMC培养上清液IL-2、IFN-γ水平显著升高(t=9.768,P=0.000;t=10.496,P=0.000;t=22.504,P=0.000;t=23.204,P=0.000),IL-4和IL-10水平显著降低(t=-18.659,P=0.000;t=-19.495,P=0.000;t=-26.258,P=0.000;t=-27.402,P=0.000)。猪带绦虫重组BCG-TSOL18疫苗可诱导仔猪产生Th1型的保护性免疫应答,从而抵抗猪带绦虫卵的感染攻击。
To study the protective immunity induced by egg challenge of Taenia solium pigs infected with recombinant BCG-TSOL18 vaccine of Taenia solium. Twelve 40-day-old healthy piglets were divided into three groups: 1011 CFU recombinant BCG-TSOL18 vaccine gavage group, BCG control group and PBS control group. Immunization every 2 weeks, a total of 2 immunizations. Four weeks after the last immunization, the eggs were challenged with Taenia solium eggs. After March, the piglets were dissected and the cysticerci were separated to calculate the rate of shrinkage. The anterior vena cava blood was collected and the serum was collected and prepared for peripheral blood monouclear cell, PBMC). Serum IgG, IgG1 and IgG2a levels and the levels of IL-2, IFN-γ, IL-4 and IL-10 in PBMC culture supernatants were detected by ELISA. The proliferation of PBMCs was detected by MTT assay. Recombinant BCG-TSOL18 vaccine immunized piglets with challenged Taenia solium eggs obtained 79.44% protection. Serum IgG and IgG2a levels were significantly elevated in the recombinant BCG-TSOL18 vaccine group compared with BCG and PBS control groups (t = 13.026, P = 0.000; t = 13.253, P = 0.000; (T = 14.199, P = 0.000), and the level of IgG1 was significantly decreased (t = -9.118, P = 0.000; t = -10.372, P = 0.000) = 0.000). The levels of IL-2 and IFN-γin PBMC culture supernatants were significantly increased (t = 9.768, P = 0.000; t = 10.496, P = 0.000; t = 22.504, P = 0.000; = 0.000), IL-4 and IL-10 levels were significantly lower (t = -18.659, P = 0.000; t = -19.495, P = 0.000; t = -26.258, P = 0.000; ). The recombinant BCG-TSOL18 vaccine of Taenia solium can induce a protective Th1-type immune response in piglets and thus resist the infection of Taenia solium eggs.
