目的和方法:应用核仁组成区相关嗜银蛋白(AgNORs)染色结合图像分析技术,观察皮质类固醇激素地塞米松对体外培养的NIH/3T3成纤维细胞AgNORs增生的抑制作用。结果:(1)地塞米松对正常和不同因子刺激的成纤维细胞AgNORs的增生均显示不同程度的抑制作用,其抑制强弱的顺序为:正常>5-羟色胺>IL-1和肝素;(2)地塞米松的抑制作用与其浓度密切相关,对正常、5-羟色胺和肝素、IL-1刺激的成纤维细胞增生的最佳抑制浓度分别为50、100和150×103μg/L。结论:上述实验结果为进一步研究地塞米松抗纤维化作用以及抑制成纤维细胞增生的作用机制提供了重要依据。
Purpose and Methods: The inhibitory effects of corticosteroid dexamethasone on the proliferation of AgNORs in NIH / 3T3 fibroblasts were observed by AgNORs staining and image analysis. Results: (1) Dexamethasone inhibited the proliferation of fibroblasts fibroblasts stimulated by normal and different factors to various degrees, the order of inhibition was: normal> serotonin> IL-1 and heparin; ( 2) The inhibitory effect of dexamethasone is closely related to the concentration of dexamethasone. The optimal inhibitory concentration of normal, serotonin and heparin, IL-1-stimulated fibroblast proliferation were 50,100 and 150 × 103μg / L, respectively. Conclusion: The above results provide an important basis for further study on the antifibrotic effect of dexamethasone and the mechanism of inhibiting fibroblast proliferation.
