目的:研究厚朴叶中多糖的超声提取最佳提取工艺,同时将粗多糖脱蛋白后经处理得到的多糖进行抗氧化性实验,检测其抗氧化活性。方法:设计正交实验进行超声法提取厚朴叶中的多糖,采用苯酚-硫酸法对厚朴叶多糖含量进行测定。利用Fenton反应原理,测定厚朴叶多糖清除羟自由基活性能力。结果:超声提取厚朴叶多糖的最佳工艺条件为:提取温度为50℃,提取时间60min,液料比为20:1,溶液pH为8,此时厚朴叶多糖得率为25.03mg/g。抗氧化性实验显示厚朴叶多糖对羟自由基的清除作用显著,厚朴叶多糖的半抑制浓度IC50值为0.08g/L。结论:该方法简便、科学,为厚朴叶多糖提取工艺提供了理论基础。厚朴叶多糖具有较强的抗氧化性活性,可为厚朴药材资源的综合利用开发提供科学依据。
OBJECTIVE: To study the optimum extraction technology of polysaccharides from Magnolia officinalis leaf by ultrasonic extraction. At the same time, the polysaccharide obtained after deproteinization of crude polysaccharide was tested for its antioxidant activity and its antioxidant activity was tested. Methods: Orthogonal experiment was designed to extract polysaccharides from Magnolia officinalis by ultrasonic method. The polysaccharide content of Magnolia officinalis was determined by phenol - sulfuric acid method. Using Fenton reaction principle, the activity of hydroxyl radical scavenging activity of Magnolia biloba leaf polysaccharide was determined. Results: The optimal extraction conditions were as follows: the extraction temperature was 50 ℃, the extraction time was 60 min, the ratio of liquid to material was 20: 1, the pH of the solution was 8, the yield of polysaccharide from Magnolia officinalis was 25.03 mg / g. Antioxidant experiments showed that the Magnolia bark polysaccharide scavenging effect on hydroxyl free radicals significantly, Magnolia leaf polysaccharide half-inhibitory concentration IC50 value of 0.08g / L. Conclusion: This method is simple and scientific, providing a theoretical basis for the extraction of polysaccharides from Magnolia Bark. Magnolia biloba polysaccharide has strong antioxidant activity, which can provide a scientific basis for the comprehensive utilization of Magnolia officinalis resources.
