目的:探讨增殖细胞核抗原(PCNA)和天冬氨酸特异性半胱氨酸蛋白酶-3(Caspase-3)在肺癌中的表达及其临床意义。方法:应用免疫组化SP法对80例肺癌病例和相对应的80例正常肺组织进行PCNA蛋白和Caspase-3蛋白半定量分析。结果:PCNA蛋白在肺癌组织中阳性表达率为65.0%,Caspase-3在肺癌组织中阳性表达率为32.5%,与相应正常肺组织中的阳性表达率有显著差异(P<0.05)。PCNA蛋白的表达与淋巴结转移、分期和分化程度明显相关(P<0.05),Caspase-3蛋白表达与肺癌的组织病理学类型、淋巴结转移以及分化程度明显相关(P<0.05),与病理分期却未表现出相关性(P>0.05),PCNA蛋白与Caspase-3蛋白的表达呈负相关,这其中PCNA(+)Caspase-3(-)的肺癌群体低/未分化危险性是非PCNA(+)Caspase-3(-)的27倍(OR=27.0,P<0.05)。结论:肺癌组织PCNA蛋白表达增强和Caspase-3蛋白表达减弱,两者的表达与肺癌的病理类型,淋巴结转移,分期和分化程度具有一定关系,关于这两种蛋白的研究对临床工作有一定指导意义。
Objective: To investigate the expression of proliferating cell nuclear antigen (PCNA) and caspase-3 in lung cancer and its clinical significance. Methods: Semi-quantitative analysis of PCNA protein and Caspase-3 protein in 80 cases of lung cancer and 80 cases of normal lung tissues were performed by immunohistochemical SP method. Results: The positive expression rate of PCNA protein in lung cancer tissues was 65.0%. The positive expression rate of Caspase - 3 in lung cancer tissues was 32.5%, which was significantly different from that in normal lung tissues (P <0.05). The expression of PCNA protein was significantly correlated with lymph node metastasis, staging and differentiation (P <0.05). Caspase-3 protein expression was significantly correlated with histopathological types, lymph node metastasis and differentiation of lung cancer (P <0.05) There was a negative correlation between the expression of PCNA protein and Caspase-3 protein in PCNA (+) Caspase-3 (-) lung cancer group. The low / undifferentiated risk of PCNA (+) Caspase- 27-fold (OR = 27.0, P <0.05) for Caspase-3 (-). CONCLUSIONS: The expression of PCNA protein and the expression of Caspase-3 in lung cancer tissues are weakened. The expression of them is related to the pathological type, lymph node metastasis, staging and differentiation degree of lung cancer. The research of these two proteins has some guidance on clinical work significance.
