目的:从信号转导通路上重要调节因子cAMP(环磷酸腺苷),PKA(蛋白激酶A)及PKC(蛋白激酶C)的角度,探讨抑郁大鼠模型受体后信号转导的变化及加味逍遥汤的干预作用。方法:将SD大鼠随机分为6个组,每组10只,即正常组、模型组、氯米帕明组(13.5 mg.kg-1)、加味逍遥汤高、中、低剂量组(生药11.7,5.85,2.925 g.kg-1)。造模同时各组按10 mL.kg-1体重给药,正常组、模型组给予等量的生理盐水,每日1次,连续24 d。末次给药后1 h,快速断头处死,在冰上剥离大脑,分离海马迅速放入冻存管,投入液氮中,随后置于-70℃低温冰箱保存备用。按ELISA试剂盒说明用酶标仪检测大鼠海马cAMP,PKA及PKC的含量变化。结果:与模型组比较,各组均能增加大鼠海马cAMP,PKA及PKC的含量,差异有显著性意义(P<0.05),加味逍遥汤各剂量组cAMP,PKA及PKC的含量增加,随剂量增加作用加强,与模型组比较差异有显著性意义(P<0.05);加味逍遥汤高、中剂量组与氯米帕明组相比cAMP,PKA及PKC含量差异没有显著性意义。加味逍遥汤高、中剂量组与低剂量组相比,差异有显著性意义(P<0.05)。结论:加味逍遥汤可能是通过调节受体后cAMP,PKA及PKC信号传导通路发挥抗抑郁作用。
OBJECTIVE: To investigate the changes of signal transduction and receptor signaling in depressed rat models from the perspectives of cAMP (cyclic AMP), PKA (protein kinase A) and PKC (protein kinase C) in signal transduction pathways Happy soup intervention. Methods: SD rats were randomly divided into 6 groups, 10 rats in each group, namely normal group, model group, clomipramine group (13.5 mg.kg-1), Jiawei Xiaoyao Decoction high, medium and low dose group Crude drug 11.7, 5.85, 2.925 g.kg-1). At the same time, each group was given body weight 10 mL.kg-1. The normal group and model group were given the same amount of saline once a day for 24 consecutive days. 1 h after the last administration, the rats were quickly decapitated, and the brain was peeled off on ice. The hippocampus was quickly placed in the cryopreservation tube, put into liquid nitrogen, and then placed in -70 ℃ low temperature refrigerator for future use. The contents of cAMP, PKA and PKC in hippocampus of rats were detected by ELISA kit by ELISA kit. Results: Compared with the model group, the contents of cAMP, PKA and PKC in the hippocampus of rats in each group were significantly increased (P <0.05), while the contents of cAMP, PKA and PKC in each group of Jiawei Xiaoyao Decoction increased (P <0.05). There was no significant difference in the content of cAMP, PKA and PKC between Jiaxiao Xiaoyao decoction group and clomipramine group. Jiawei Xiaoyao Tang high dose group and low dose group, the difference was significant (P <0.05). Conclusion: Jiawei Xiaoyao decoction may play an antidepressant effect by regulating cAMP, PKA and PKC signal transduction pathways.
