[目的]Klotho基因是一种与衰老相关的基因,相关研究发现,人类G-395A、F352V以及C370S位点的多态性是冠心病的独立危险因素,但尚无研究证实这3个位点同低密度脂蛋白胆固醇(low density lipoprotein-choles-terol;LDL-C)水平是否有关。本研究旨在探讨Klotho基因的以上3个多态性位点与外周血中低密度脂蛋白胆固醇水平的关系。[方法]从不同低密度脂蛋白胆固醇水平的患者的外周血中提取DNA,TaqMan探针法检测其G-395A、F352V以及C370S位点的多态性。[结果]Klotho基因G395A和F352V多态性同LDL-C水平有相关性(P﹤0.01,χ2=82.268和P﹤0.01,χ2=21.957);C370S位点的多态性与血清LDL-C水平无相关性(P=0.813,χ2=1.576)。[结论]Klotho基因G395A和F352V的多态性和血清LDL-C水平相关,而与C370S位点的多态性与血清LDL-C水平无关,G395A和F352V多态性有可能作为LDL-C升高的预测指标。
[Objective] Klotho gene is an aging-related gene. Related studies have found that human G-395A, F352V and C370S polymorphisms are independent risk factors for coronary heart disease. However, no studies have confirmed these three sites With low density lipoprotein cholesterol (low density lipoprotein-choles-terol (LDL-C) levels are related. The purpose of this study was to investigate the relationship between the above three polymorphisms of Klotho gene and the level of LDL cholesterol in peripheral blood. [Methods] DNA was extracted from the peripheral blood of patients with different LDL cholesterol levels. The polymorphisms of G-395A, F352V and C370S were detected by TaqMan probe. [Results] The polymorphisms of Klotho gene G395A and F352V were correlated with the level of LDL-C (P <0.01, χ2 = 82.268 and P <0.01, χ2 = 21.957) No correlation (P = 0.813, χ2 = 1.576). [Conclusion] The polymorphisms of Klotho gene G395A and F352V are correlated with serum LDL-C level, but not with the C370S polymorphism and serum LDL-C level. The G395A and F352V polymorphisms may be used as LDL-C level High prediction index.
