为准确测定血浆中尼莫地平浓度,本文建立了离子抑制性反相高效液相色谱法。血浆样品碱化后以乙醚-氯仿混合溶剂提取,提取液挥干,甲醇溶解后进样分析。采用μ-BoudapakC_(18)径向加压柱系统,甲醇-磷酸盐缓冲液为流动相,紫外检测波长为236nm。选择尼群地平为内标,峰面积定量。在10~200ng/ml内线性关系良好(r=0.9987),方法稳定可靠。
In order to accurately determine the concentration of nimodipine in plasma, ion-suppressed reversed-phase high performance liquid chromatography was established in this paper. The plasma samples were alkalified and extracted with ether-chloroform mixed solvent. The extract was evaporated to dryness and dissolved in methanol for injection analysis. The μ-BoudapakC_ (18) radial column system was used. The mobile phase consisted of methanol-phosphate buffer and the UV detection wavelength was 236 nm. Select nitrendipine as internal standard, peak area quantitative. In 10 ~ 200ng / ml within a good linear relationship (r = 0.9987), the method is stable and reliable.
