The protoplast culture of Gramineae is still a difficult problem in plant cell and tissue culture. The results of somatic embryo formation and mature plant regeneration from cultured protoplasts of Polypogon fugax Nees ex Steud are reported. Regenerated plantlets have been obtained from suspension culture protoplasts and regenerated mature plants from protoplasts directly isolated with embryogenic calli. The latter offered an example of new approach to protoplast culture of Gramineae. A discussion of culture media and culture methods is also presented.
