目的 :探索贯叶连翘外植体组织培养的适合培养基配方。方法 :将贯叶连翘种子发芽产生的根、带节茎段、叶作为外植体 ,培养在含不同种类及浓度激素的 Murashige and Skoog ( MS)基本培养基上 ,诱导愈伤组织、芽和根的形成 ,并进行试管苗的移栽。结果 :不同激素配比和不同种类外植体对愈伤组织和芽的形成有较大区别。 2 ,4-二氯苯氧乙酸 ( 2 ,4- D)有利于促进贯叶连翘愈伤组织的形成 ;在细胞分裂素中 ,6 -苄基氨基嘌呤 ( 6 - BA)促进生芽的作用很明显 ,而激动素 ( KT)、玉米素 ( ZT)则不明显 ;α-萘乙酸 ( NAA) 1mg/ L 与 6 - BA0 .2 m g/ L 的组合有利于贯叶连翘的生根。 结论 :含有 2 ,4- D的 MS培养基有利于愈伤的诱导 ,外植体根最适合丛生芽的快速诱导。
Objective : To explore the suitable medium formulation for tissue culture of Hypericum perforatum explants. METHODS: Roots, stem segments and leaves from the seeds of St. John’s wort were germinated as explants and cultured on Murashige and Skoog (MS) medium containing different hormones and concentrations to induce callus, buds and roots. The formation of transplanted plantlets. RESULTS: Different hormone combinations and different types of explants had significant differences in the formation of callus and shoots. 2 ,4-Dichlorophenoxyacetic acid (2,4-D) promotes the formation of Hypericum perforatum callus; in cytokinins, 6-benzylaminopurine (6-BA) promotes budding. Obviously, kinetin (KT) and zeatin (ZT) were not obvious; the combination of α-naphthylacetic acid (NAA) 1 mg/L and 6-BA 0.2 mg/L was beneficial to the rooting of Hypericum perforatum. Conclusion : MS medium containing 2,4-D is beneficial to the callus induction. The explant root is most suitable for the rapid induction of clustered shoots.
