目的:探讨Syk基因在子宫内膜良性及恶性病变中的表达及其在子宫内膜癌发生、发展中的可能作用。方法:分别采用逆转录-聚合酶链反应及蛋白质印迹法检测52例子宫内膜癌、30例子宫内膜不典型增生、28例复杂型增生、30例单纯型增生和40例正常子宫内膜组织中Syk基因及蛋白的表达。结果:从正常子宫内膜组、不典型增生组至子宫内膜癌组中Syk基因和蛋白表达依次降低,差异有统计学意义,P<0.05;Syk基因和蛋白在复杂型增生、单纯型增生及正常子宫内膜中的表达差异无统计学意义,P>0.05;子宫内膜癌中,Syk基因和蛋白的阳性表达与临床分期(P<0.05)、淋巴结转移(P<0.01)以及肌层浸润深度(P<0.05)明显相关,而与组织学分级无明显相关性,P>0.05。结论:Syk低表达或缺失可能与子宫内膜癌的发生、浸润及转移有关。
Objective: To investigate the expression of Syk gene in benign and malignant endometrial lesions and its possible role in the development and progression of endometrial carcinoma. Methods: 52 cases of endometrial carcinoma, 30 cases of endometrial dysplasia, 28 cases of complex hyperplasia, 30 cases of simple hyperplasia and 40 cases of normal endometrium were detected by reverse transcription - polymerase chain reaction and Western blot, respectively Syk gene and protein expression in tissues. Results: The expression of Syk gene and protein in normal endometrium group, atypical hyperplasia group and endometrial carcinoma group decreased in turn, the difference was statistically significant (P <0.05). The Syk gene and protein in complex hyperplasia, simple hyperplasia (P <0.05), lymph node metastasis (P <0.01) and the expression of Syk gene and protein in endometrial carcinoma (P <0.05), but not in normal endometrium Depth of invasion (P <0.05), but no significant correlation with histological grade, P> 0.05. Conclusion: The low expression or deletion of Syk may be related to the occurrence, invasion and metastasis of endometrial carcinoma.
