目的 建立大鼠 C6脑胶质瘤模型 ,行一般观察和MRI检查以确定颅内肿瘤生长情况 ,寻找简易判定颅内肿瘤大小的可靠指标 .方法 体外培养大鼠 C6胶质瘤细胞 ,应用立体定向法将含 10 g· L- 1 琼脂糖和 1× 10 6 C6细胞悬液 10μL注入大鼠右脑尾状核 .接种后行一般观察和 MRI检查 ,对5组接种大鼠分别在第 10 ,15 ,2 0 ,2 5日和自然死亡前做主动脉多聚甲醛灌注固定 ,对脑组织和肿瘤标本进行切片观察和HE染色 .结果 5 0只接种大鼠均有脑内肿瘤生长 (10 0 % ) ,远处和颅外转移率为 0~ 4% .结论 成功建立大鼠 C6脑胶质瘤模型 ,接种后脑内肿瘤呈球状生长 ,很少出现颅外转移 .大鼠的生存时间可作为判定颅内肿瘤生长大小的指标 .
OBJECTIVE: To establish a rat model of C6 glioma and to observe the growth of intracranial tumors by MRI and MRI.Methods Rat C6 glioma cells were cultured in vitro and stereotactically Law 10g · L-1 agarose and 1 × 10 6 C6 cell suspension 10μL injected into the right cerebellar caudate nucleus of rats.After inoculation, the general observation and MRI examination were carried out on the 5 groups of inoculated rats at the 10th, 15, 20, 25, and aorta paraformaldehyde perfusion fixation before and after natural death were performed to observe the brain tissue and tumor specimens and HE staining.Results All of the 50 vaccinated rats had intracerebral tumor growth (100% ), Distance and extracranial metastasis rate was 0 ~ 4% .Conclusion The successful establishment of rat C6 glioma model, after inoculation of tumors in the brain spherical growth, with little extracranial metastasis.The survival time of rats can be used as a judge Intracranial tumor growth size indicators.
