目的和方法:为了解SD大鼠肝卵圆细胞体外转化过程中胞浆游离钙及cAMP含量的变化。用荧光探针标记法检测胞浆游离钙浓度。以放射免疫法检测cAMP含量。结果:从喂饲3’-甲基-N,N-二甲基-4-氨基偶氯苯(3’-Me-DAB)大鼠分离到的卵圆细胞,体外生长至30代时发生转化,生长速度加快。80%的转化卵圆细胞为异倍体,并能在软琼脂中呈克隆性生长。尽管细胞在转化过程中未见表皮生长因子受体含量改变,但细胞游离钙升高和cAMP水平明显降低,反映细胞传导系统发生了改变。结论:这种细胞第二信使物质的改变使细胞保持恶性转化表型并能不断生长。
Purpose and Methods: To investigate the changes of cytosolic free calcium and cAMP during the in vitro transformation of hepatic oval cells of SD rats. Cytoplasmic free calcium concentration was detected by fluorescent probe labeling. Radioimmunoassay detection of cAMP content. RESULTS: Oval cells isolated from rats fed 3’-methyl-N, N-dimethyl-4-amino-chlorobenzene (3’-Me-DAB) Grow faster. Eighty percent of transformed oval cells are aneuploid and grow clonally in soft agar. Although no change of epidermal growth factor receptor was observed in the cells during transformation, the increase of free calcium and cAMP level in cells showed that the cell conduction system had changed. Conclusion: The changes of the second messenger substance in this cell make the cell maintain the malignant transformed phenotype and grow continuously.
