目的 :应用光镜及免疫组织化学技术观察转基因糖尿病小鼠颌下腺的形态学改变以及血管内皮生长因子 (Vascularendothelialcellgrowthfactor ,VEGF)在颌下腺表达的变化 ,旨在说明糖尿病时颌下腺功能损害与上述因子间的关系。方法 :引进日本C5 7BL/ksj—db/ +m表型正常隐性基因小鼠 ,近亲交配 ,其纯合子后代 ,即为db/db(单基因遗传自然发病型 )糖尿病小鼠。取 3、4、6、8、10月龄db/db糖尿病小鼠及相应年龄段的db/ +m正常小鼠 (对照组 )颌下腺 ,常规固定包埋 ,切片 ,行HE及SP免疫组化染色 ,图象分析仪计数颌下腺内VEGF免疫阳性的细胞数。结果 :随着糖尿病进展 ,颌下腺组织萎缩 ,但间质增生 ,可见血管增多。在db/db糖尿病小鼠VEGF阳性细胞数随着糖尿病进展有一明显增多趋势 ,且与各同龄对照组相比明显增强 (p <0 .0 5 )。结论 :VEGF在糖尿病颌下腺表达增强,可能与缺氧有关 ,进而导致间质血管增生 ,新血管形成 ,这也是糖尿病性微血管病变的相关因素之一。
OBJECTIVE: To observe the morphological changes of submandibular gland and the expression of vascular endothelial growth factor (VEGF) in the submandibular gland of mice with transgenic diabetes mellitus and immunohistochemistry by light microscopy and immunohistochemistry to illustrate the relationship between the submandibular gland dysfunction and the above factors during diabetes mellitus . Methods: The mice with the normal phenotype of C5 7BL / ksj-db / + m were introduced and their offspring were mated. The homozygous offspring were db / db diabetic mice with a single gene inheritance. Submandibular glands of db / db diabetic mice at 3, 4, 6, 8 and 10 months old and normal db / + m mice of the corresponding age group (control group) were routinely fixed and sectioned and subjected to HE and SP immunohistochemistry Staining, image analyzer counting submandibular gland VEGF immunoreactive cells. Results: With the progress of diabetes, submandibular gland atrophy, but interstitial hyperplasia, visible blood vessels. The number of VEGF-positive cells in db / db diabetic mice showed a significant increase trend with the progression of diabetes mellitus, and was significantly increased compared with control group of the same age (p <0.05). Conclusion: The increased expression of VEGF in diabetic submandibular glands may be related to hypoxia, leading to interstitial vascular hyperplasia and neovascularization, which is also one of the related factors of diabetic microangiopathy.
