目的:研究高良姜总黄酮(TFAO)的超声萃取工艺及其体外抗氧化作用。方法:采用正交设计优化TFAO的超声萃取工艺;采用化学模拟体系测定其抑制脂质体过氧化及清除·OH、O2g和DPPH·的能力;采用体外试验研究TFAO对小鼠肝、脑匀浆中GSH-Px活力、H2O2诱导的MDA生成和红细胞溶血的影响。结果:TFAO超声萃取的最佳条件:萃取溶剂为50%乙醇,超声时间45min,料液比1∶20,最高得率48.17mg/g。TFAO对自由基的清除能力较强,其清除·OH、O2g、DPPH·和抑制脂质体过氧化的IC50分别为152、258、7.11、22.44mg/L;可增强GSH-Px活力,抑制MDA的产生和红细胞(RBC)溶血,其中等浓度时效果与VC相当。结论:TFAO具有较强的抗氧化作用。
Objective: To study the ultrasonic extraction process of TFAO and its anti-oxidative effect in vitro. Methods: The orthogonal design was used to optimize the ultrasonic extraction process of TFAO. The ability of inhibiting lipid peroxidation and scavenging of OH, O2g and DPPH · was determined by chemical simulation system.
