目的研究肠型胃癌不同发展阶段基因表达谱的改变。方法应用基因芯片技术对3例肠型胃癌不同发展阶段基因表达特征进行检测分析。基因芯片包括576个cDNA克隆,其中包括506个靶克隆,51个是本课题组前期工作经消减杂交得到的克隆,另外455个克隆则根据它们在肿瘤中的作用而选取。应用Cluster和TreeView软件进行聚类分析,研究各种基因与胃癌发生发展不同阶段的关系。结果经基因芯片技术分析得到了181个差异表达基因,至少在胃癌发生发展的一个阶段其Cy5∶Cy3的比值大于2或者小于0 .5。其中48个基因上调,133个基因下调。聚类分析将差异表达基因分成了5个特征性的组,反映了不同阶段的基因表达特征。同时发现一些重要的差异表达基因, SEC23IP、LIPF、EST(BQ291520)、SLC5A1、PG、CXCR4、DICER1、SH3GL2以及IGF2R等。结论认识并发现了胃癌差异基因表达特征以及一些重要基因,对肠型胃癌发生发展及转移的进一步研究将具有重要作用。
Objective To study the changes of gene expression profiles in different stages of intestinal-type gastric cancer. Methods The gene expression profiles of three types of intestinal-type gastric cancer at different developmental stages were detected by gene chip technique. The cDNA microarray consisted of 576 cDNA clones, including 506 target clones, and 51 clones obtained by subtractive hybridization in our previous work. The other 455 clones were selected based on their role in the tumor. Cluster and TreeView software were used to analyze the relationship between various genes and different stages of gastric cancer. Results 181 differentially expressed genes were obtained by gene chip analysis. The ratio of Cy5: Cy3 was greater than 2 or less than 0.5 at least during the development of gastric cancer. Among them, 48 genes were up-regulated and 133 genes were down-regulated. Clustering analysis differentially expressed genes into five characteristic groups, reflecting the characteristics of gene expression at different stages. Some important differentially expressed genes, SEC23IP, LIPF, EST (BQ291520), SLC5A1, PG, CXCR4, DICER1, SH3GL2 and IGF2R were also found. Conclusion Recognizing and discovering the differentially expressed genes of gastric cancer and some important genes, further study on the development and metastasis of enteric gastric cancer will play an important role.
