目的 了解Bcl 2对HSV 1感染原代培养神经元的调节作用。方法 用流式细胞术(FCM)和Westernblot(WB)方法检测正常组、病毒组、山梨醇组及山梨醇加病毒组在不同时间、不同条件下Bcl 2变化情况。结果 病毒组较正常组、山梨醇加病毒组较山梨醇组Bcl 2蛋白表达上调 ,且与病毒毒力有关。正常组随培养时间延长 ,Bcl 2蛋白表达含量降低。结论 HSV 1感染培养小鼠皮层神经元时 ,Bcl 2蛋白表达上调 ,阻止细胞凋亡并延长细胞寿命。
Objective To investigate the regulatory effect of Bcl 2 on primary cultured HSV-1-infected neurons. Methods The changes of Bcl 2 in normal group, virus group, sorbitol group and sorbitol plus virus group at different time and different conditions were detected by flow cytometry (FCM) and Western blot (WB). Results The Bcl 2 protein expression in the virus group was higher than that in the normal group and sorbitol plus the virus group compared with the sorbitol group, and was related to the virus virulence. The normal group with the incubation time, Bcl 2 protein expression decreased. Conclusions HSV 1 can up-regulate the expression of Bcl 2 protein in cultured cortical neurons, prevent apoptosis and prolong cell life.
