培养基的成份小球藻干粉20.0,葡萄糖20.0,食盐5.0,琼脂40.0,蒸馏水加至1,000。制法Ⅰ液:小球藻干粉10 g,木瓜素0.8 g,pH 7.0的缓衡磷酸盐水800ml,混合置45℃温箱中24小时。Ⅱ液:小球藻干粉10g,胃蛋白酶粉10g,1 N盐酸80ml,混合置45℃温箱中24小时。将Ⅰ液与Ⅱ液混和于2,000 ml容量的烧瓶中,再置于45℃温箱中6小时,滴定pH7.0,加入葡萄糖、食盐、琼脂,以蒸馏水补足至1,000ml,煮沸至琼脂完全融解,不用再较正酸碱度,立即放于45℃温箱中趁热使自行沉淀
