目的探讨半胱氨酸对玻璃化卵子冷冻后发育潜能及纺锤体正常形态率的研究。方法在玻璃化冷冻卵子中添加1 mmol/L半胱氨酸,观察其发育潜能。同时利用纺锤体观察仪计算纺锤体正常形态率。最后选取部分卵子固定染色后,经激光共聚焦显微镜观察其形态变化。结果 1 mmol/L半胱氨酸能有效提高玻璃化卵子冷冻后受精率(70.8%和84.8%)(P<0.05),但对3-4细胞和桑葚+囊胚获得率无明显改善作用(P>0.05)。同时半胱氨酸提高了复苏后卵子纺锤体正常形态率(53.3%和74.4%)(P<0.05)。结论玻璃化冷冻引起卵子发育潜能下降,异常纺锤体增加。半胱氨酸可以有效提高玻璃化冷冻卵子受精率及纺锤体正常形态率。
Objective To investigate the developmental potential of cysteine after freezing vitrified ovum and the normal morphology of spindle. Methods 1 mmol / L cysteine was added into the vitrified frozen eggs to observe its developmental potential. At the same time using the spindle observer to calculate the spindle normal morphology rate. Finally selected part of the egg fixed staining, observed by laser confocal microscopy morphological changes. Results 1 mmol / L cysteine could effectively improve the fertilization rate (70.8% and 84.8%) of vitrified eggs (P <0.05), but had no significant effect on the yield of 3-4 cells and mulberry + blastocysts P> 0.05). At the same time, cysteine increased the normal morphological rate of ovum spindle (53.3% and 74.4%) after resuscitation (P <0.05). Conclusion Vitrification caused a decrease in egg developmental potential and an increase in abnormal spindle. Cysteine can effectively improve the fertilization rate of vitrified frozen eggs and spindle normal morphology rate.
