目的探讨中药复方治疗抗生素相关性腹泻(antibiotic associated diarrhea,AAD)的药理机制。方法应用荧光定量PCR技术检测2种中药复方(参苓白术散和理中汤)对2株艰难梭菌临床分离株(A+B+和A-B+)体外培养下毒素基因表达的影响。结果在A+B+艰难梭菌中,参苓白术散组对tcd A和tcd B基因的表达均有显著或是完全抑制,理中汤也有显著的抑制;而对于A-B+艰难梭菌,2种中药复方则完全抑制tcd B基因的表达。结论通过影响毒素基因的表达可能是中药复方治疗AAD的药理学机制之一。
Objective To investigate the pharmacological mechanism of traditional Chinese medicine compound in the treatment of antibiotic associated diarrhea (AAD). Methods Fluorescent quantitative PCR was used to detect the effects of two traditional Chinese medicines (Shenling Baizhu San and Li Zhong Tang) on toxin gene expression in two strains of clinical isolates of C. difficile (A + B + and A-B +) in vitro. Results In the A + B + Clostridium difficile, the expression of tcd A and tcd B genes was significantly or completely inhibited by Shenling Baizhu San, while Li Zhong Tang was also significantly inhibited; while for A-B + C. difficile 2 Traditional Chinese medicine compound completely inhibits the expression of tcd B gene. Conclusion One of the pharmacological mechanisms of traditional Chinese medicine compound in the treatment of AAD may be through the influence of toxin gene expression.
