目的探讨抵抗素对胰岛β细胞胰-十二指肠同源盒因子-1(PDX-1)的表达、胰岛素分泌及细胞增殖、凋亡的影响。方法βTc6细胞分为对照(NC)组和抵抗素组(浓度20~200ng/ml),干预6、12、24h,CCK-8法测定细胞增殖能力,DNA Ladder检测细胞凋亡,RT-PCR检测PDX-1基因表达,放射免疫法测定胰岛素分泌。结果 (1)与NC组比较,20、40ng/ml抵抗素组各干预时段,胰岛β细胞的PDX-1表达、增殖活力、凋亡、胰岛素分泌差异均无统计学意义(P>0.05);(2)随抵抗素浓度在60~200ng/ml内增加,干预时间由6~24h延长,细胞PDX-1表达、增殖活力、胰岛素分泌均呈现浓度和时间依赖性降低(P均<0.05),且细胞凋亡条带逐渐明显。结论在一定的浓度和时间范围内,抵抗素可呈浓度和时间依赖性下调胰岛β细胞PDX-1表达,损害胰岛素分泌,抑制细胞增殖、诱导其凋亡。
Objective To investigate the effects of resistin on the expression of PDX-1, insulin secretion and cell proliferation and apoptosis in pancreatic β cells. Methods βTc6 cells were divided into control group (NC) and resistin group (concentration 20 ~ 200ng / ml) for 6, 12 and 24 hours. Cell proliferation was determined by CCK-8 assay. Apoptosis was detected by DNA Ladder. PDX-1 gene expression, radioimmunoassay for insulin secretion. Results (1) Compared with the NC group, there was no significant difference in the expression of PDX-1, proliferation, apoptosis and insulin secretion between the groups of 20 and 40 ng / ml resistin at each time point (P> 0.05). (2) With the increase of resistin concentration in 60 ~ 200 ng / ml, the intervention time prolonged from 6 to 24 h, the concentration of PDX-1 and the activity of proliferation and insulin secretion decreased in concentration and time dependent manner (all P <0.05) And apoptotic bands gradually obvious. Conclusion Within a certain concentration and time range, resistin can down-regulate the expression of PDX-1 in pancreatic β cells in a concentration- and time-dependent manner, impairing insulin secretion, inhibiting cell proliferation and inducing apoptosis.
