目的:分析卵泡刺激素受体(Follicle stimulating hormone receptor,FSHR)胞外区蛋白的二级结构,在线预测FSHR胞外区潜在的B细胞表位。方法:利用DNAStarProtein软件对FSHR胞外区蛋白的二级结构和表面特性进行分析,联合在线B细胞表位预测,预测FSHR胞外区蛋白可能的抗原表位。合成预测的B细胞表位肽,对其抗原性进行鉴定。结果:FSHR胞外区蛋白可能的抗原表位区域为17~34、42~44、116~122、142~147、179~193、239~241、266~270、279~282、288~307。选择4个区域合成肽段表位区域能与免疫抗血清发生抗原特异性反应。结论:应用多参数成功预测了FSHR胞外区蛋白抗原的B细胞表位。
OBJECTIVE: To analyze the secondary structure of the extracellular domain of follicle stimulating hormone receptor (FSHR) and to predict the potential B cell epitopes in the extracellular domain of FSHR. Methods: The secondary structure and surface properties of extracellular domain of FSHR were analyzed by DNAStarProtein software. Combined with online B cell epitope prediction, the potential epitopes of extracellular domain of FSHR were predicted. The predicted B-cell epitope peptide was synthesized and its antigenicity was identified. RESULTS: The possible epitopes of extracellular domain of FSHR were 17 ~ 34,42 ~ 44,116 ~ 122,142 ~ 147,179 ~ 193,239 ~ 241,266 ~ 270,279 ~ 282,288 ~ 307. Select the four regions of synthetic peptide epitopes with the immune antiserum antigen-specific reaction. Conclusion: B-cell epitopes of FSHR extracellular domain protein antigen were successfully predicted by using multiple parameters.
