钙离子载体A23187可以引起细胞内Ca2+浓度升高,并可诱导某些细胞程序死亡。本文发现1g/mlA23187作用于HL-60细胞4小时即可诱导程序死亡,以无毒性浓度的蛋白磷酸酶2B(PP2B)的抑制剂环抱菌素A(CsA)(0.5-3g/m1)预处理可以抑制A23187诱导的HL-60细胞程序死亡,磷酸酶1、2A、2C的抑制剂okadaicacid(OA)和酪氨酸磷酸酶的抑制剂钒酸钠(sodiumorthovanadate,SoV)则无此效应。流式细胞术测定群体细胞内总Ca2+变化发现,CsA不抑制A23187诱导的胞内Ca2+浓度升高,表明CsA作用于Ca2+升高后的下游事件。
Calcium ionophore A23187 can cause elevated intracellular Ca2 + concentration and induce some programmed cell death. It was found that 1g / ml A23187 could induce programmed death in HL-60 cells for 4 hours, and cyclosporin A (CsA) (0.5-3g / ml) could be induced by non-toxic inhibitor of protein phosphatase 2B (PP2B) Pretreatment could inhibit apoptosis induced by A23187 in HL-60 cells. No effect was found for okadaicacid (OA), an inhibitor of phosphatase 1,2A, 2C and sodiumorthovanadate (SoV), a tyrosine phosphatase inhibitor. The results of flow cytometry showed that CsA did not inhibit the increase of intracellular Ca2 + concentration induced by A23187, indicating that CsA acts on the downstream events of Ca2 + upregulation.
