目的 探讨洛伐他汀 (Lovastatin)影响MCF 7乳腺癌细胞增殖功能的可能机制。方法 采用生长曲线 ,NBT还原实验 ,逆转录 PCR检测RbmRNA和westernblotting分析Rb蛋白表达等实验技术。 结果 不同剂量的Lovas tatin均可抑制MCF 7细胞增殖能力并促进细胞的分化 ;Lovastatin处理 4 8h和 72h后RbmRNA较未处理组细胞表达增强 ;处理 4 8h后 pRb蛋白表达随剂量的增加而逐渐减弱 ,而 72h后各处理组pRb蛋白表达均明显低于未处理组 ,且同剂量组间处理 72hpRb蛋白表达明显低于 4 8h的处理组。 结论 Lovastatin可能通过作用于细胞周期进程中RbmRNA合成及其蛋白的表达 ,来促进MCF 7乳腺癌细胞的分化
Objective To investigate the possible mechanism of Lovastatin on the proliferation of MCF-7 breast cancer cells. Methods The experimental techniques such as growth curve, NBT reduction assay, RT-PCR detection of Rb mRNA and western blotting analysis of Rb protein expression were performed. Results Different doses of Lovas tatin could inhibit the proliferation of MCF 7 cells and promote the differentiation of cells. After 48 h and 72 h of Lovastatin treatment, the expression of Rb mRNA increased compared with that of untreated cells. The expression of pRb protein decreased gradually with the increase of dose , While the protein expression of pRb in each treatment group was significantly lower than that of the untreated group after 72 hours. The expression of 72hpRb protein in the same dosage group was significantly lower than that in the 48 h treatment group. Conclusion Lovastatin may promote the differentiation of MCF-7 breast cancer cells through the role of Rb mRNA in cell cycle progression and its protein expression
