目的:探讨毒素清颗粒对支原体肺炎老龄大鼠免疫功能和细胞因子的影响。方法:SD大鼠随机分为青年对照组、青年模型组,老龄对照组、老龄模型组、毒素清组和阿奇霉素组,采用气管插管法复制支原体肺炎模型。分别以毒素清(3.995 g.kg-1.d-1)、阿奇霉素(23 mg.kg-1.d-1)灌胃7 d。第8天取材,检测外周血白细胞(WBC)和中性粒细胞(NEU)计数,支气管肺泡灌洗液(BALF)支原体PCR和分泌性免疫球蛋白(sIg)A、纤维连接蛋白(FN)表达,肺脏含水量和病理,血清肿瘤坏死因子(TNF)-α、白介素(IL)-6、IL-8表达。结果:阿奇霉素组BALF肺炎支原体阳性为0例(0%),毒素清组阳性3例(30%)。青年和老龄模型组BALF中sIgA和FN分别较青年、老龄对照组显著降低(P<0.01),肺组织含水量、外周血TNF-α,IL-6,IL-8显著升高(P<0.01),且较青年模型组变化显著(P<0.01)。与老龄模型组比较,毒素清组和阿奇霉素组sIgA,FN显著升高(P<0.01),肺组织含水量、TNF-α,IL-6,IL-8显著降低(P<0.01);毒素清组BALF中sIgA明显高于阿奇霉素组(P<0.01),IL-8低于阿奇霉素组(P<0.05)。结论:毒素清对老龄大鼠支原体肺炎作用明显,其机制可能与增强免疫功能、降低炎症因子表达,减轻炎症反应有关。
OBJECTIVE: To investigate the effects of toxin granule on immune function and cytokines in aged rats with mycoplasma pneumonia. Methods: SD rats were randomly divided into young control group, youth model group, aging control group, aging model group, toxin group and azithromycin group. Tracheal intubation was used to replicate mycoplasma pneumonia model. The rats were treated with toxin (3.995 g.kg-1.d-1) and azithromycin (23 mg.kg-1.d-1) respectively for 7 days. On day 8, WBC and neutrophil count (NEU), Mycoplasma pneumoniae (BAL) mycoplasma PCR, sIgA and fibronectin (FN) , Lung water content and pathology, serum tumor necrosis factor (TNF) -α, interleukin (IL) -6 and IL-8. Results: 0 cases were positive for Mycoplasma pneumoniae in azithromycin group and 3 cases were positive in toxin group (30%). SIgA and FN in BALF in young and aged model group were significantly lower than those in young and old control group (P <0.01), and the content of lung tissue, TNF-α, IL-6 and IL-8 in peripheral blood were significantly increased ), And significantly changed than the young model group (P <0.01). The levels of sIgA and FN in the toxin group and azithromycin group were significantly higher than those in the model group (P <0.01), and the contents of water, TNF-α, IL-6 and IL- The sIgA levels in BALF were significantly higher than those in azithromycin group (P <0.01), IL-8 was lower than that in azithromycin group (P <0.05). Conclusion: Toxin Qing obvious effect on mycoplasma pneumonia in aged rats, its mechanism may be related to enhancing immune function, reducing the expression of inflammatory cytokines and reducing the inflammatory reaction.
