目的:观察限制饮食对运动小鼠生长发育、运动能力和免疫功能的影响,以及益气养血方的中药调理作用。方法:将38只4周龄雌性Balb/c小鼠随机分为正常进食组、模型组及服药组。采用转棒式疲劳仪进行递增强度跑步训练,三组动物运动量相同。正常组不限制饮食,模型组在运动训练同时限制饮食,服药组在运动训练及限制饮食同时服用益气养血方药,实验期1个月。实验结束后,记录小鼠运动成绩,检测其外周血RBC和Hb含量,称量胸腺和脾脏以及子宫湿重并计算脏器指数,以MTT法测定脾脏T淋巴细胞增殖能力和ELISA方法检测脾细胞IL-2、IFN-γ含量。结果:实验过程中正常组小鼠体重保持增长,模型组小鼠在造模过程中几乎停止生长,而服药组小鼠能维持体重缓慢增长。模型组小鼠的运动成绩和脏器指数、红细胞计数与血红蛋白含量均显著低于正常组。各组小鼠的脾脏淋巴细胞增殖能力和脾细胞分泌IL-2、IFN-γ能力未见显著差异。结论:益气养血方对限制饮食运动小鼠的血色素水平下降,生长发育延迟与运动能力下降、免疫功能损害有一定的保护作用。
OBJECTIVE: To observe the effects of diet restriction on the growth and development, exercise capacity and immune function in mice and the conditioning effect of traditional Chinese medicine. Methods: Thirty-eight female Balb / c mice of 4 weeks old were randomly divided into normal feeding group, model group and medication group. The use of rotating rod-type fatigue device for increasing strength running training, the same amount of animal movement in the three groups. The normal group did not limit the diet. The model group also restricted the diet during exercise training. The medication group took the Yiqi Yangxue recipe at the same time as the exercise training and diet restriction. The experiment period was 1 month. At the end of the experiment, the motor performance of mice was recorded, the contents of RBC and Hb in peripheral blood were measured, the thymus and spleen and the wet weight of uterus were weighed and the organ index was calculated. The proliferation of T lymphocytes in spleen was measured by MTT assay and spleen cells IL-2, IFN-γ content. Results: The body weight of mice in normal group kept increasing during the experiment. Mice in model group almost stopped growing during the process of modeling, but the mice in treatment group maintained a slow growth of body weight. Mice in model group had significantly lower scores of motor performance, organ index, red blood cell count and hemoglobin than those in normal group. The spleen lymphocyte proliferation ability and splenocyte secretion of IL-2 and IFN-γin the mice of all groups showed no significant difference. Conclusion: Yiqi Yangxue Prescription can decrease the level of hemoglobin, delay the growth and development, decrease the exercise capacity and impair the immune function in the diet-induced mice.
