目的:通过观察养精种玉汤对过多雄激素诱导卵泡胰岛素抵抗的影响,初步探讨其促卵泡发育的机制。方法:体外培养猪卵泡,经过多雄激素作用48 h后,检测猪卵泡对葡萄糖的摄取量和卵泡培养液上清残糖量。分别检测正常组、抵抗组和复方组卵泡胰岛素受体底物-1(IRS-1)和葡萄糖转运体4(GLUT4)蛋白表达水平以及卵泡培养液上清残糖量。结果:过多雄激素作用48 h后,卵泡对葡萄糖的摄取量较正常组下降39.56%(P<0.01),卵泡培养液上清残糖较正常组升高29.08%(P<0.01)。当卵泡胰岛素抵抗时,IRS-1与GLUT4蛋白表达量较正常组分别降低74%与94.56%(P<0.01);加入养精种玉汤复方改善胰岛素抵抗后,复方组IRS-1与GLUT4蛋白表达量较抵抗组分别升高2倍与5倍(P<0.01),卵泡培养液上清残糖较抵抗组降低33.6%(P<0.001)。结论:过多雄激素能诱发卵泡胰岛素抵抗,养精种玉汤能改善卵泡胰岛素抵抗,其机制与IRS-1与GLUT4表达量有关。
OBJECTIVE: To observe the effect of Jingjing Zhongyu Decoction on insulin resistance induced by excessive androgen in follicles, and to explore its mechanism of follicular development. Methods: Porcine follicles were cultured in vitro, and the amount of glucose uptake and the amount of residual carbohydrate in the supernatant of follicles were detected after 48 hours of androgen treatment. The expression of insulin receptor substrate-1 (IRS-1) and glucose transporter 4 (GLUT4) in follicles of normal group, resistance group and compound group were detected respectively, as well as the residual carbohydrate content of follicle culture supernatant. Results: The amount of glucose uptake in follicles decreased by 39.56% (P <0.01) and the residual carbohydrate content in follicle culture supernatant increased by 29.08% (P <0.01) compared with the normal group after 48 hours of treatment with excessive androgen. When follicular insulin resistance, IRS-1 and GLUT4 protein expression decreased by 74% and 94.56% (P <0.01) respectively compared with the normal group; after adding Jingjing Zhongyu decoction to improve insulin resistance, IRS-1 and GLUT4 protein Compared with the resistant group, the residual amount of residual carbohydrate in the supernatant of the follicle culture medium decreased by 33.6% (P <0.001). Conclusion: Too much androgen can induce insulin resistance in follicles, and Yangjing Decoction can improve insulin resistance in follicles. The mechanism is related to the expression of IRS-1 and GLUT4.
